摘要
目的:以人脂肪间充质干细胞(human adipose -derived stem cells,hADSCs)为种子细胞,体外构建hADSCs 膜片,分析细胞膜片的生物学特点,为进一步进行动物实验奠定基础。方法胶原酶消化法分离纯化hADSCs,观察其增殖、生长特性和组织学形态;体外构建 hADSCs 膜片,膜片成熟后倒置显微镜、苏木精-伊红染色对膜片进行形态学检测。结果hADSCs 为长梭形,形态均一,增殖活跃。体外培养28天后获得半透明膜状细胞膜片,有弹性,倒置显微镜下观察细胞复层生长,细胞间紧密连接。苏木精-伊红染色证实脂肪干细胞膜片是一种由多层细胞和细胞外基质组成的聚集体。结论hADSCs 可体外成功构建细胞膜片,细胞数量及活性较高。
Objective To use human adipose -derived stem cells (hADSCs) as the seeding cells to construct the hADSCs membrane, then explore its biological characteristics for the purpose of the further graft experiment.Methods hADSCs were isolated and purified by collagenase digestion and were used to its proliferation, morphology and growth characteristics.After the construction of hADSCs sheet in vitro, invert microscope and H -E staining were employed for morphological observation of the sheet.Results hADSCs has a long spindle shape, uniform morphology and high prolif-eration.After 28 days in vitro culture of hADSCs, a translucent and elastic membranaceous sheet was formed.hADSCs grew in multilayer observed under inverted microscope and intercellular tight junctions.H -E staining confirmed that adipose derived stem cell membrane was aggregates composed of multilayer cells and extracellular matrix.Conclusion The cell -sheet constructed successfully by hADSCs has a high cell number and activity.
