生姜种植土壤中螺螨酯降解菌Enterobacter sp.QD26-6的筛选与鉴定 被引量:5

Screening and Identification of Spirodiclofen-degrading Bacteria Enterobacter sp. QD26-6 for Soil under Ginger Cultivation

摘要 [目的]利用生物降解治理螺螨酯污染土壤。[方法]采用富集培养和高效液相色谱测定等方法,从山东青岛生姜种植地采集的土壤样品中筛选出1株螺螨酯高效降解菌。对该菌株进行了菌种鉴定并测定了农药降解谱。[结果]筛选出的菌株QD26-6能够以螺螨酯为唯一碳源,在无机盐培养基中培养120 h后对100 mg/L螺螨酯的降解率为71.72%。该菌株在培养72 h后对吡虫啉和三氟羧草醚的降解率为33.74%~39.14%;对乙嘧酚磺酸酯、丁醚脲和烯酰吗啉的降解率为21.93%~26.60%。根据菌落形态、革兰氏染色结果及16S rDNA序列分析,初步鉴定该菌株为肠杆菌属(Enterobacter sp.)。[结论]菌株QD26-6对环境中的螺螨酯生物修复具有很好的应用潜力。 [Aims] In order to remediate spirodiclofen-contaminated soil through biodegradation. [Methods] A high-effective spirodiclofen-degrading bacteria was isolated from ginger-cultivated soil in Qingdao by enrichment culture and HPLC method. The strain identification was studied and the pesticides degrading spectrum was also determined. [Results] The isolated strain QD26-6 could use spirodiclofen as the sole carbon source, the degradation rate of spirodiclofen(100 mg/L) was 71.72% in MSM after 120 h cultivation. As the strain was cultivated for 72 h, the degradation rates of imidacloprid and acifluorfene were 33.74-39.14%, the degradation rates of bupirimate, diafenthiuron and dimethomorph were 21.93-26.60%. Based on colonies morphology, gram staining result and 16 S rD NA gene sequence analysis, the strain was identified as Enterobacter sp. [Conclusions] The strain QD26-6 showed good application potential for bio-remediation of spirodiclofen-contaminated environment.
