摘要
目的检测成骨-破骨细胞共育体系中应用加味三根汤含药血清后抗酒石酸酸性磷酸酶(TRACP)以及白细胞介素-1β(IL-1β)、肿瘤坏死因子(TNF-α)的变化,探讨加味三根汤的作用机制。方法建立成骨-破骨细胞共育体系,制作SD大鼠含药血清,分为空白对照组,加味三根汤低、中、高剂量组,每组设6个复孔,采用酶联免疫吸附试验(ELISA)检测共培养体系上清液中TRACP及IL-1β、TNF-α的含量。结果经鉴定成功培养成骨细胞和破骨细胞,并建立成骨-破骨细胞共育体系。加味三根汤含药血清中、高剂量组TRACP活性低于空白血清对照组,差异具有统计学意义(P<0.05)。与空白血清对照组比较,加味三根汤中、高剂量组IL-1β含量有所下降,加味三根汤高剂量组TNF-α含量降低,差异具有统计学意义(P<0.05)。结论加味三根汤能够抑制共育体系中破骨细胞的功能,此作用可能是通过抑制IL-1β、TNF-α的分泌而达到。
Objective To detect the changes of tartrate-resistant acid phosphatase (TRACP), IL-1 [3 and TNF-c~ in osteoblasts-osteoclasts co-culture system and to discuss its mechanism. Methods The osteoblasts-osteoclasts co-culture system model was established and SD rats were randomized into blank contol group, "Jiawei Sangen Decoction" groups. Six pores were set up in each group. The expressions of TRACP, IL-1β and TNF-α were detected by ELISA. Results The osteoblasts and osteoclasts were cultured and osteoblasts-osteoclasts co-culture system was set up. The TRACP in the medium and high dose "Jiawei Sangen Decoction" group were lower than that of the control group (P 〈 0.05 ). Compared with the blank group, the IL- 1β level in the medium and high dose "Jiawei Sangen Decoction" groups were reduced, and the TNF-α in the high dose "Jiawei Sangen Decoction" group was reduced significantly ( P 〈 0.05 ). Conclusion "Jiawei Sangen Decoction" can inhibit the function of osteoclasts in osteoblasts-osteoclasts co-culture system, its mechanism may be associated with inhibiting the secretion of IL- 1β and TNF-a.
