HPLC法同时测定心脑脉通胶囊中葛根素与蓝萼甲素的含量

Simultaneous Determination of Puerarin and Glaucocalyxin A in Xinnaomaitong Capsules by HPLC

目的建立HPLC法同时测定心脑脉通胶囊中葛根有效成分葛根素及蓝萼香茶菜中有效成分蓝萼甲素的含量。方法采用Diamonsil C18色谱柱（250 mm×4.6 mm,5μm）;流动相为乙腈（A）-0.5%冰醋酸水溶液（B）,梯度洗脱;流速1.0 ml·min-1;检测波长240 nm。结果葛根素、蓝萼甲素分别在21.9~109.5、5.04~50.4μg·ml-1范围内线性关系良好;平均回收率分别为98.0%、98.7%;RSD分别为1.4%、1.0%。结论本方法可靠稳定,可用于心脑脉通中葛根素、蓝萼甲素的含量测定。

Objective To develop an HPLC method for simultaneous determination of puerarin and glaucocalyxin A in Xinnaomaitong capsules.Methods Diamonsil C18column（250 mm × 4.6 mm,5 μm） was used.The mobile phase was methanol（A） and 0.5% acetic water solution（B） in a gradient elution mode at the flow rate of1.0 ml·min-1.The detection wavelength was 240 nm.Results The content of puerarin showed a good linearity within the range of 21.9-109.5 μg·ml-1.The average recovery was 98.0%（RSD = 1.4%）.The content of glaucocalyxin A showed a good linearity within the range of 5.04-50.4 μg·ml-1.The average recovery was 98.7%（RSD = 1.0%）.Conclusion The developed method is accurate with high reproducibility and stability,which can facilitate quality control of Xinnaomaitong capsules.
