摘要
目的建立HPLC法测定止咳合剂中吗啡的含量。方法色谱柱:Wonda Sil C18(4.6 mm×150 mm,5μm);流动相:0.01 mol·L-1磷酸二氢钾溶液-0.0025 mol·L-1庚烷磺酸钠溶液(含0.1%三乙胺,磷酸调p H至2.5)-乙腈(90∶10);流速:1.0 ml·min-1;检测波长:220 nm;柱温:30℃;进样量10μl。结果吗啡在0.525~42.0μg·ml-1范围内线性关系良好(r=1.0000),平均回收率为99.97%,RSD为1.27%(n=9)。结论本方法简便准确,重复性好,能够用于止咳合剂中吗啡的含量测定和质量控制。
Objective To establish an HPLC method for determination of morphine in anticough mixture.Methods Wonda Sil C18(4.6 mm × 150 mm,5 μm) was used.The mobile phase consisted of 0.01 mol·L-1KH2PO4-0.0025 mol·L-1C7H15 Na O3(0.1% triethylamine,adjusted to p H 2.5 with phosphoric acid)-C2H3N(90∶ 10);at a flow rate of 1.0 ml·min-1.The detection wavelength was set at 220 nm.The temperature of the column was 30 ℃,and the volume of injection was 10 μl.Results A good linear correlation of morphine was observed within the range of 0.525-42.0 μg·ml-1(r = 1.0000).The average recovery was 99.97% with an RSD of1.27%(n = 9).Conclusion This method is simple,accurate and quick,which can be used for content determination of morphine and quality control of anticough mixture with good selectivity and repeatability.
