北衙金矿中蜡样芽孢杆菌的检测

摘要 利用地质体表土壤中蜡样芽孢杆菌数量作为指示菌去探寻隐覆金矿是一种非常有效的方法.采用可培养法、定性PCR扩增法及荧光原位杂交法对滇西北北衙金矿矿区蜡样芽孢杆菌的分布情况进行了检测.结果表明:选用蜡样芽孢杆菌专用培养基-卵黄琼脂培养基没有分离到该菌;定性PCR方法也未能扩增到蜡样芽孢杆菌的特异性解旋酶gyrB基因;采用荧光原位杂交的方法也未检测到该菌株,故推断北衙金矿区不存在蜡样芽孢杆菌异常现象.因此,以蜡样芽孢杆菌作为指示菌,寻找覆隐金矿的方法并不一定适用于所有金矿. The Bacillus cereus in geological surface soil has been reported as a very effective method to indicate the blind gold deposit.Culturable method,qualitative PCR amplification and fluorescence in situ hybridization are used to determine the distribution of Bacillus cereus in Beiya gold deposit located in southwest China.The results show that Bacillus cereus has not been isolated from the special culture medium of yolk agar medium.The specificity helicase gyrB gene also fails to be amplified by qualitative PCR methods,and it can not be detected by using fluorescence in situ hybridization method.It is,therefore,inferred that there is not Bacillus cereus anomalies in Beiya gold deposit.Therefore,as an indicator bacterium,Bacillus cereus indicating blind gold deposit is not necessarily applicable to all gold mines.
