摘要
本文主要目的是为了观察消渴通脉胶囊原料灌胃给药的小鼠急性毒性反应。经预试确定按最大给药量法设计试验,小鼠40只,♂♀各半,随机分为给药组和阴性对照组。给药组小鼠单次灌胃给予18.75g/kg的消渴通脉胶囊原料,阴性对照组给予等容量的蒸馏水,给药容量均为30mL/kg,24h内每隔6h连续给药3次,累积剂量为56.25g/kg。给药后自由饮食饮水,连续14d观察小鼠体质量、行为活动以及死亡情况。给药组小鼠观察期间内体质量及一般状况无明显异常变化,无死亡例数。试验结果显示,在本试验条件下,消渴通脉胶囊原料的最大给药量为56.25g/kg,相当于原生药203.85g,该剂量相当于人临床剂量的102倍。
To study the effect of Xiaoke Tongmai capsule crude drug on acute toxicity in mice after intragastric administration. The maximum quantity was determined according to the design of pilot test. Forty mice were randomly divided into the treatment group and negative control group. For the treatment group, Xiaoke Tongmai capsule crude drug, with 18.75 g/kg as maximum drug dose and 30 mL/kg as maximum drug volume,is administrated three times in 24 h,while for the negative control group, distilled water is adopted. The accumulated dose was 56.25 g/kg. After administration, mice were free to forage and drink for 2 weeks,and the bodyweight,behavior and death were recorded. No abnormality in bodyweight and behavioral activity was observed in the treatment group. No death was discovered. Under this experimental condiion,the dose of 56.25 g/kg is the maximum dose of Xiaoke Tongmai capsule crude drug. It was about 203.85 g crude drug and 102 times of the clinic dose.
