高效液相色谱法考察木绣球不同部位中绿原酸含量

摘要 为比较木绣球不同部位中绿原酸的含量,采用索氏提取法提取木绣球花、花托、茎、叶等四个部位的绿原酸并用HPLC法测定含量。色谱条件如下:Eclipse XDB—C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈-0.1%磷酸水溶液(10:90),检测波长为327mm,流速为1.0mL/min。在上述色谱条件下,绿原酸在0.34—6.80μg/mL范围内线性良好。结果表明木绣球不同部位中绿原酸含量为:花托(1.14%)〉花(1.02%)〉茎(0.51%)〉叶(0.45%)。木绣球中花托部位绿原酸含量最高,木绣球整体均具有潜在的研究与开发价值。 The contents of chlorogenic acid in flowers, torus, stems and leaves of Viburnum macrocephalum was determined by HPLC. V. macrocephalum was extracted by Soxhlet extraction method. The chromatographic condition was established as follows:Ecllpse XDB-C18 (4.5 mm × 250 mm,5 μm) column,the mobile phase composed of acetonitrile- 0.1% phosphoric acid aqueous solution ( 10: 90) ,the detection wavelength was 327 nm,the flow rate was 1.0 mL/min. Under the developed HPLC conditions ,good linear relationship was established within the range of 0.34-6.80 μg/mL. The content of chlorogenic acid in different parts of V. macrocephalum was in following order:toms (1.14%) 〉 flowers ( 1.02% ) 〉 stems (0.51%) 〉 leaves (0.45%). The torus of V. macrocephalum had the highest content of chlorogenic acid. V macrocephalum had the potential exploitation value.
