摘要 目的分析甘草异黄酮2'-羟化酶(Gu I2'H)c DNA序列及其编码的蛋白质的结构特征。方法以Gu I2'H c DNA序列为研究对象,应用生物信息学软件和网络工具分析其c DNA序列及其编码的蛋白质的结构、功能及分子进化特征。结果 Gu I2'H c DNA全长2 425 bp,含有1 521 bp的开放读码框(ORF),编码538个氨基酸残基的多肽。该蛋白质为跨内质网膜蛋白,与其他物种的I2'H具有高度的序列同源性,与同科植物黄芩、大豆的I2'H亲缘关系较近。结论甘草异黄酮2'-羟化酶c DNA编码异黄酮2'-羟化酶,属于细胞色素P450 81E家族成员。 Objective To analyze the structure characteristics of an isoflavone 2'-hydroxylase cDNA sequence (GuI2'H) and the deduced amino acid sequence in Glycyrrhiza uralensis. Methods The complete GuI2'H cDNA sequence and the structure, function, phylogeny of the deduced amino acid sequence were predicted by bioinformatics software and network tools. Results The complete cDNA sequence of GuI2'H was 2 425 bp with an open reading frame of 1 521 bp,encoding a polypeptide of 475 amino acid residues. GuI2'H was predicted to be an endoplasmic reticulum transmembrane protein, which has a high sequence identity with I2'H of other species, and closely related to I2'H in Astragalus mongholicus, Glycine max and other Leguminosae plants. Conclusion GuI2'H encodes a protein of isoflavone 2'-hydroxylase, which belongs to the cytochrome P450 81E family.
