摘要
以亨氏马尾藻(S.henslowianum)为原料,提取分离纯化后的岩藻聚糖硫酸酯组分SPF3,采用β-葡聚糖酶对其进行可控酶降解,并分别就加酶量、反应温度、酶解时间对岩藻聚糖硫酸酯降解的影响进行考察,通过L9(34)正交试验对SPF3进行酶法降解,得到最佳降解工艺为加酶量0.03%,反应温度50℃,酶解时间1 h,p H值6.0,多糖质量浓度1 mg/m L。通过对降解前后岩藻聚糖硫酸酯的总糖、硫酸根、相对分子质量进行分析,表明可以在较短的时间内使岩藻聚糖硫酸酯多糖分子量降低。
The project intends to adopt modern biochemical separation to separate and puriy fucoidan SPF3 from Sargassum henslowianum C. Agardh. Using-glucanase enzyme to degrade SPF3. By L9(34)orthogonal experiment design,the optimum processing conditions of degration are determined. The enzyme quantity,the reaction temperature,the enzyme degeneration time three factors as the independent variable,the best degeneration condition to add the enzyme quantity is 0.03%,the temperature for50 ℃,the enzyme degeneration time is 1 h,p H 6.0,sugar density 1 mg/m L. Through to analyze degrade total sugar,sulfate,the relative molecular,indicated that may cause the fucoidan molecular weight to reduce in such a short time.
