参龙汤对脑缺血大鼠脑一氧化氮合酶活性及其表达影响

Effect of Shenlong Tang on the Activity and Expression of Nitric Oxide Synthase in Cortex with Cerebral Ischemia Rats

摘要 目的:观察参龙汤对脑缺血大鼠大脑皮层中一氧化氮(NO)含量、诱导型一氧化氮合酶(i-NOS)活性及其蛋白表达的影响。方法:利用双侧颈总动脉结扎制备慢性脑缺血的模型,50只Wistar大鼠分为:假手术组,脑缺血模型组,脑缺血加脑复康组,脑缺血加参龙汤大、小剂量组。造模以及药物处理4周后处死大鼠,用ELISA法检测各组大鼠的大脑皮层中NO含量、i-NOS活性,用免疫组织化学染色法观察大鼠大脑皮层脑组织i-NOS蛋白的表达。结果:脑缺血模型组大鼠NO含量、i-NOS活性升高;大剂量参龙汤干预后NO含量、i-NOS活性明显降低,与假手术组比,差异显著(P<0.05),模型组大鼠海马NOS蛋白表达显著增加;参龙汤大剂量给药后海马NOS蛋白表达阳性细胞数明显减少,与模型组比有显著性差异(P<0.05)。结论:参龙汤能降低脑缺血大鼠大脑皮层中NO的含量、i-NOS活性,减少慢性脑缺血大鼠大脑皮层NOS蛋白的表达。 Objective : To investigate the effect of Shenlong Tang ( SLT ) decoction on the level of nitric oxide ( NO ) and activity of nitric oxide synthase ( NOS ) and NOS expression in cortex of cerebral ischaemia rats. Methods: 50 rats were randomly divided into 5 groups: control group with slam operation; cerebral ischemia model group ; positive control group of pisacetam; SLT groups ( low and high dosage ). The cerebral ischemia rats' model was established by two-vessel occlusion. After modeling was administrating with SLT and pisacetam for 28 days, the contents of NO and inducible NOS in cerebral cortex were measured, protein expression in the cerebral cortex was detected. Results : The levels of NO and activity of NOS in the cerebral cortex was increased significantly in the model group, compared with slam operation group there was significant difference, P〈0.05. High dosage SLT and pisacetam could significantly increase the levels of NO and activity of NOS in the cerebral cortex, there was significant difference compared with model group, P〈0.05. The positive cell nmnber of NOS expression in cerebral cortex area was significantly decreased in SLT high dosage group, compared with model group there was significant difference, P〈0.05, Conclusion : SLT could decrease the levels of NO and activity of NOS in the cerebral cortex and decrease the expression of NOS.
