RP-HPLC with DAD Switch Determination of Hydroxysafflor Yellow A and Ferulic acid in Dieda Huoxue Powders

目的：建立同时测定跌打活血散中羟基红花黄色素A和阿魏酸含量的方法。方法：采用RP HPLC法,Waters Symmetry C18色谱柱（250 mm×4.6 mm,5μm）,以甲醇∶0.2%甲酸水溶液（45∶55）为流动相;体积流量1.0 m L·min^-1;柱温30℃;检测波长为403 nm（0-11 min）;324 nm（11-20 min）。结果：羟基红花黄色素A和阿魏酸分别在0.248-2.48μg、0.08-0.80μg质量浓度范围内与其峰面积均呈良好的线性关系（r≥0.999 9）;平均加样回收率均大于95.0%。结论：建立的方法快速、准确,可用于活血跌打散的质量控制。

Objective：To establish an RP HPLC method for simultaneously determining two components in Dieda Huoxue Powders,including hydroxysafflor yellow A and ferulic acid. Methods：The determination was performed on a Waters Symmetry C18（250 mm×4.6 mm,5 μm）,the mobile phase consisted of methanol-0.2% formic acid at a flow rate of 1.0 m L/min with 30 ℃. DAD was used as the detector. The detection wavelengths were 403 nm（0-11 min）and 324 nm（11-20 min）. Results：The method has a good linearity in the ranges of 0.248-2.48 μg for hydroxysafflor yellow A and 0.08-0.80 μg for ferulic acid,respectively（r ≥ 0.999 9）. The average recoveries of the two compoments were more than 95.0%. Conclusion：This method with strong specificity is simple,accurate and can be used for quality control of Dieda Huoxue Powders.
