摘要
采用乙醇回流法提取丹参、赤芍等药材,加入辅料(蔗糖∶糊精=2∶1)制备冠心颗粒,采用RP-HPLC法测定颗粒的有效成分芍药苷、丹参素、丹酚酸B和丹酚酸A,以Agilent TC-C18(150 mm ×4.6 mm,5μm)为分析柱,以甲醇-0.2%磷酸溶液为流动相梯度洗脱,流速1.0 mL/min,柱温28℃。测得芍药苷、丹参素、丹酚酸B、丹酚酸A分别在5.44~87.04,6.68~106.88,9.48~151.68,3.06~48.96μg/mL内线性良好,r≥0.9992。
Guanxin particles were prepared by adding different proportions sucrose and dextrin (2∶1) into the extractives from ethanol backflow extraction .The effective components such as paeoniflorin ,Danshen-su,salvianolic acid B and A were determined using RP-HPLC.The Agilent TC-C18 (150 mm ×4.6 mm, 5 μm) column was used,with methanol-0.2%phosphoric acid solution as mobile phase and gradient elu-tion.The flow rate was 1.0 mL/min at 28 ℃.The linear relationships were good in the range of 5.44~87.04 μg/mL for paeoniflorin,6.68~106.88 μg/mL for Danshensu,9.48~151.68 μg/mL for salvian-olic acid B and 3.06~48.96μg/mL for salvianolic acid A,respectively.The correlation coefficients were no less than 0.999 2.
