摘要
采用乙醇提取和液液萃取的方法对短柄小连翘茎枝进行了提取。通过硅胶柱色谱、Sephadex LH-20柱色谱、反相中压柱色谱和高效液相等多种色谱方法相结合,从短柄小连翘乙醇提取物的乙酸乙酯部位中分离得到了7个双环氧木脂素化合物。经MS和NMR等波谱技术分别鉴定为松脂醇(pinoresinol,1),5’-甲氧基-松脂素(medioresinol,2),8-乙酰基松脂醇(8-ace-toxypinoresinol,3),表松脂醇(epipinoresinol,4),(+)-丁香树脂醇[(+)-syringaresinol(5)]、(+)-1-羟基丁香树脂醇[(+)-1-hydmxysyringaresin01(6)]和erythro-buddlenolE(7)。化合物1~7均为首次从该植物中分离得到。抗氧化活性筛选结果显示,化合物7在1×10^-6mol·L^-1浓度下,对Fe^2+-Cys诱导大鼠肝微粒体丙二醛(MDA)生成的抑制率为38%,而相同浓度下,阳性对照药Vit E的抑制率为35%,表明化合物7具有显著的抗氧化作用。
The stems and branches of Hypericum petiolulatum were extracted by alcohol and liquid-liquid extraction. Seven furo- furanlignans were isolated from the ethyl acetate fraction of ethanol extract of H. petiolulatum by using silica gelchromatography, Sepha- dex LH-20 chromatography, medium-pressure liquid chromatography and preparative HPLC. Their structures were identified by the spectroscopic methods as pinoresinol( 1 ) , medioresinol ( 2 ) , 8-acetoxypinoresinol ( 3 ) , epipinoresinol ( 4 ) , ( + ) -syringaresinol ( 5 ) , ( + )-1-hydroxysyringaresinol(6) and erythro-buddlenolE (7). All the isolates were firstly found in H. petiolulatum. In the bioassay, compound 7 showed remarkable antioxidative activity inhibiting Fe +2-cystine induced rat liver microsomal lipid peroxidation with inhibi- tory rate 38% at a concentration of 1 × 10 ^-6 mol · L^-1 ( positive control Vit E with the inhibitory rate of 35% at the same concentra- tion).
