四逆散对慢性应激大鼠空间学习记忆能力的影响 被引量:4

摘要 目的:观察四逆散对慢性应激大鼠空间学习记忆能力的影响。方法:选取SD大鼠40只,随机分为正常对照组、应激模型组、四逆散低剂量组和四逆散高剂量组,每组10只。建立慢性心理应激动物模型,正常对照组给予0.9%氯化钠溶液2mL·d-1灌胃;应激模型组给予0.9%氯化钠溶液2mL·d-1灌胃;四逆散低剂量组给予四逆散低浓度药液2mL·d-1灌胃;四逆散高剂量组给予四逆散高浓度药液2mL·d-1灌胃。采用Morris水迷宫实验检测大鼠空间学习记忆能力。结果:定位航行实验结果示,各组大鼠逃避潜伏期组间存在显著差异(P<0.01);与正常对照组比较,应激模型组、四逆散低剂量组和四逆散高剂量组逃避潜伏期明显延长(P<0.01,P<0.05);与应激模型组比较,四逆散低剂量组逃避潜伏期明显缩短(P<0.05)。空间探索实验结果示,入水后前20s,与正常对照组比较,应激模型组及四逆散低剂量组大鼠穿越原平台区域次数明显减少(P<0.05),四逆散高剂量组无明显变化(P>0.05);与四逆散高剂量组比较,应激模型组和四逆散低剂量组明显减少(P<0.01或P<0.05)。入水120s后各组穿越原平台区域次数高低次序没有变化,仅四逆散高剂量组较四逆散低剂量组穿越原平台区域次数明显增加(P<0.05),其余各组比较差异均无统计学意义(P均>0.05)。结论:长期慢性应激刺激可引起大鼠空间学习记忆能力损伤,导致空间学习记忆障碍;四逆散可减轻慢性应激对大鼠空间学习记忆能力的损伤,提高慢性应激大鼠空间学习和记忆能力;不同剂量的四逆散对慢性应激大鼠空间学习和记忆能力的保护作用也有变化。 Objective:To observe the effects of Frigid Extremities Powder on spatial Learning and memory abilities of chronic stress rats. Methods:SD rats were randomly divided into the normal group, the chronic stress group, the low dosage group and the high dosage group, with 10 rats in each group. The rats models of chronic stress were created and rats in the low dosage group and the high dosage group were given respectively with the low and high concentration of Frigid Extremities Powder. The learning and memory abilities of rats were examined by Morris Water Maze. Results : In the place navigation test, there was the statistical signifi- cance among groups ( P 〈 0.01 ). Compared with the normal gnmp, escape latency of rats were prolonged obviously in the chronic stress group,the low dosage group and the high dosage group (P 〈 0.01,P 〈 0.05). The escape latency of rats in the low dosage group was shorter than normal group( P 〈 0.05 ). In the first 20 seconds of the spatial probe test, compared with the normal group, platform crossing times (times of crossing the former site of removed hidden platform) was significantly reduced in the chronic stress group, the low dosage group, and no obvious change in the high dosage group. Compared with the high dosage group, plat- form crossing times was significantly reduced in the chronic stress group and the low dosage group (P 〈0.01 ,P 〈0.05 ). In the 120 seconds of the spatial probe test,the order of platform crossing times was same as it did before. The platform crossing times of the high dosage group was more than that of the low dosage group. Beyond that, there was no obvious difference among groups ( P 〉 0.05 ). Conclusion : ①Chronic stress impairs spatial learning and memory abilities of stress rats. ②Frigid Extremities Powder could significantly alleviate the spatial learning and memory disturbance, improve spatial learning and memory abilities of chronic stress rats. ③ Different dosage of Frigid Extremities Powder have different protective effect of spatial learning and memory abilities of chronic stress rats.
