西洋参总皂苷对心力衰竭模型大鼠心肌能量代谢的影响 被引量:2

摘要 目的:观察西洋参总皂苷(AGS)对心力衰竭模型大鼠心肌能量代谢的影响。方法:将48只SD大鼠随机分为假手术组、模型组、AGS低剂量组、AGS中剂量组、AGS高剂量组和阳性药物组,每组8只。采用冠状动脉结扎法制作SD大鼠心力衰竭模型,分别给药12周后处理动物,采用称量法测定全心质量指数(HW/BW)、左心质量指数(LVW/BW),采用生物发光法测定左心室肌三磷酸腺苷(ATP)浓度,采用荧光法测定左心室肌线粒体膜通透性转变孔道(MPTP)活性。结果:与假手术组比较,模型组的HW/BW、LVW/BW显著增加、ATP浓度显著降低,MPTP活性显著增强(P均<0.05);与模型组比较,AGS中剂量组、AGS高剂量组及阳性药物组的HW/BW、LVW/BW显著减少,ATP浓度显著升高,MPTP活性显著减弱(P<0.05)。结论:西洋参总皂苷可改善心力衰竭模型大鼠的心肌肥厚,其机制可能与西洋参总皂苷通过下调心肌线粒体MPTP活性,改善心肌能量代谢有关。 Objective :To observe the effect of American ginseng total saponius (AGS) on myocardial energy metabolism of rat model with heart failure. Methods:48 SD rats were randomized into sham operation group, model group, AGS low dosage group, AGS medium dosage group,AGS high dosage group and positive drug group,with 8 rats in each group. SD rats model with heart tailure were made via coronary artery ligation method,and the rats were handled alter being given drugs for 12 weeks. HW/BW of the whole heart and LVW/BW of the left heart were measured by weighing method. The density of ATP in left ventricular muscle was measured by bioluminescence assay. The activity of MPTP of mitochondrial membrane in the left ventricular muscle was de- tected by fluorescence method. Results: Compared with these of the cham operation group, the HW/BW of the whole heart and INW/BW of the left heart increased remarkably ,and the density of ATP model group decreased greatly (P 〈 0.05 ). Compared with model group,the HW/BW and LVW/BW of AGS medium dosage group,AGS high dosage group and positive drug group de- clined obviously,the density of ATP increased significantly, and the activity of MPTP reduced remarkably ( P 〈 0. 05 ). Conclu- sion:AGS can improve the myocardial hypertrophy of rats with heart failure,whose mechanism may be related with AGS's function of regulating MPTP of mitochondrial membrane and improving myocardial energy metabolism.
