云南省1例输入性三日疟实验室检测分析 被引量:9

摘要 目的对云南省1例输入性疟疾患者的血样进行实验室检测分析。方法采用疟原虫属特异性引物和种特异性引物对该样本进行巢式PCR检测,将PCR扩增结果进行序列测定,并对获得的序列进行Blast比对,分析其分子性状。结果使用疟原虫属特异性引物r PLU5和r PLU6对血样进行PCR检测,第一轮未扩增出预期条带;使用疟原虫种特异性引物对血样进行PCR检测,扩增出预期大小约141bp的三日疟条带,无对应大小的恶性疟、间日疟和卵形疟扩增条带产生。序列分析表明,扩增片段长度为115bp,进行Blast比对发现,与Genbank中注册编号为gb|KJ934253.1的三日疟对应部分的基因序列同源性为96%。结论通过使用巢式PCR、序列测定分析等方法,基本证实该病例为三日疟原虫感染。 Objective To detect an imported malaria case in Yunnan province. Methods The blood samples from animported malaria case was tested by nested PCR with primers specific for plasmodium genus and the sequences of theamplified products were checked in NCBI. Results The results of nested PCR detection with the primers of r PLU5 andr PLU6 specific for plasmodium-genus revealed that the sample did not produce the expected enhanced band. Then 141 bp ofPlasmodium malariae in size was produced and no corresponding bands of Plasmodium falciparum and Plasmodium vivax andPlasmodium ovale were observed after amplified with promers specific for polasmodium species. Blast analysis showed that thefragment was composed of 115 bp and had a high homology of 96% to the corresponding gene sequence of Plasmodium malariaefrom Genbank accession number gb|KJ934253.1.Conclusion Generally confirmed by nested PCR detection and blastanalysis, the case was infected with Plasmodium malariae.
