高压氧预处理对局灶性脑缺血再灌注损伤大鼠脑组织NF-κB及下游靶基因iNOS的影响 被引量:4

摘要 目的探讨高压氧预处理对脑缺血再灌注损伤大鼠脑组织核转录因子NF-κB(p65)及其下游靶基因i NOS的影响。方法将36只SD大鼠按随机数字表法分为假手术组、模型组及HBO组,每组12只。模型组和HBO组均行大脑中动脉阻塞(MCAO)术。假手术组除不插线栓外,其余步骤同模型组。HBO组大鼠提前给予连续5 d高压氧预处理,最后一次预处理后3 h再行MCAO术。大鼠脑缺血再灌注24 h后进行神经功能缺损评分,并处死取脑组织病理切片观察,用Real Time-PCR和Western blot法检测各组大鼠缺血侧脑组织NF-κB(p65)及i NOS mRNA和蛋白的表达。结果 HBO组大鼠神经功能缺损评分较模型组明显改善,差异有统计学意义(P<0.05)。HBO组NF-κB(p65)和i NOS mRNA、蛋白表达量显著低于模型组,差异有统计学意义(P<0.05)。结论高压氧预处理所诱导的NF-κB活化减少,i NOS基因转录和翻译下调,可能是其抑制炎症反应,诱发脑缺血耐受的重要保护机制之一。
