白杨素对实验性糖尿病大鼠心脏损害的保护作用及其机制

摘要 目的观察白杨素对实验性糖尿病大鼠心脏损害的保护作用,探讨其保护机制。方法一次性腹腔注射链脲佐菌素(STZ)溶液建立大鼠实验性糖尿病模型。成模大鼠随机分为模型组、白杨素低剂量组[灌胃,50 mg/(kg·d)]和白杨素高剂量组[灌胃,100 mg/(kg·d)]。每周定点测量各组体重和空腹血糖,3周后禁食12 h麻醉大鼠,取血和心脏组织,测定其丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性。HE染色观察心脏组织形态学变化。结果与模型组相比,白杨素低、高剂量组大鼠血糖显著降低,血清和心脏组织SOD、GSH-Px活性升高,MDA含量下降(P<0.05),心脏损伤程度明显减轻。结论白杨素对实验性糖尿病大鼠的心脏损害有一定保护作用,可能与其抗氧化作用相关。 Objective To investigate the cardiaprotection of chrysin for exprimental diabetic rats and its related mechanism.Methods Streptozotocin (STZ) was given to induce the diabetic rats model.The model rats were divided into the model group,chrysin low dose group [ig,50 mg/(kg·d)] and chrysin high dose group [ig,100 mg/(kg·d)].Rats weight and fasting blood glucose (FBG) were detected weekly.After 3 weeks,rats were given anesthesia after 12 h ambrosia,blood and cardiac tissue were obtained.The concentration of malondialdehyde (MDA),the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in serum and myocardium were measured.The morphology changes of heart tissue was observed with HE staining.Results Compared with the model group,in the chrysin low dose group and the chrysin high dose group,the FBG level decreased,the activities of SOD and GSH-Px increased,MDA content decreased,with statistically significant differences (P < 0.05).Conclusion Chrysin treatment has a protective effect on myocardial injury in diabetic rats,which may be related to antioxidant effect.
