睾丸特异性基因BC051628在小鼠中的表达及生物信息学分析

摘要 目的研究睾丸特异性基因BC051628在小鼠中的表达特征。方法通过分析小鼠基因表达谱芯片,发现BC051628基因特异性表达于睾丸组织。采用逆转录聚合酶链反应(RT-PCR)、实时聚合酶链反应(Real-timePCR)、蛋白免疫印迹法(WesternNot)及免疫组化等方法分析该基因在小鼠不同组织及睾丸不同发育阶段的表达特征。利用生物信息学软件对该基因进行相关生物信息学分析。结果BC051628基因在小鼠各组织中呈现睾丸特异性表达并伴随明显年龄依耐性。免疫组化染色发现BC051628蛋白主要定位于精原细胞、精母细胞及圆形精子。生物信息学分析发现BC051628基因编码蛋白氨基酸序列在人和小鼠具有高度同源性,提示该基因在哺乳动物进化过程中十分保守。结论BC051628属于睾丸特异性基因,可能在生精过程发挥重要作用。 Objective To identify the expression characteristic ofBC051628 (cDNA sequence BC051628) gene in mice. Methods A new testis-specific gene BC0.51628 was identified in mice by gene expression profile analysis. The expressions ofBC051628 in different tissues and developmental stage of testes in mice were further validated by real-time PCR, western blot and immunohistochemistry. The related bioinformation analysis were done using bioinformatics software. Results BC051628 were exclusively expressed in testes in an age-dependent manner. The protein of this gene was mainly expressed in spermatogonia, spermatocytes and round spermatids. Bioinformatics analysis showed that BC051628 protein amino acid sequences had a high degree of similarity between human and mice, which indicated that this gene was highly conserved in mammals. Conclusion BC051628 is a testis-specific gene, might play an important role in spermatogenesis.
