白斑狗鱼鱼皮中抗冻蛋白的分离纯化 被引量:2

摘要 以白斑狗鱼为研究对象,选取鱼皮组织为试验材料,通过单因素及响应面试验对鱼皮抗冻蛋白提取工艺条件进行优化。以抗冻蛋白的热滞活性(THA)作为考察指标,对提取温度、提取液的p H值、提取时间及料液比进行探讨。结果表明,在提取温度8℃、提取液p H值为8.4、提取时间2 h,料液比1∶3.8(g∶m L)的工艺条件下,模型预测的热滞活性为0.072 0℃。经验证试验,发现其热滞活性为(0.069 0±0.002 4)℃,试验值与预测值差异较小,表明模型具有一定可靠性。通过柱层析对白斑狗鱼鱼皮抗冻蛋白进行分离纯化,纯化后的抗冻蛋白达到电泳级,且其分子质量约为6 400 u,热滞活性大约为(0.050 2±0.002 2)℃。 With Esox lucius as the research object and E. lucius skin tissue as experimental material, the antifreeze protein (AFP) was isolated and pu- rified by single factor experiment and response surface methodology. Using thermal hysteresis activity (THA) of AFP as index, four factors of extraction temperature, extraction buffer pH, extraction time and solid-liquid ratio was optimized. Eventually, the results showed that the THA of AFP from skin tissues was up to 0.072 0 ℃ under the condition of extraction temperature 8 ℃, pH 8.4, extraction time 2 h and solid-liquid ratio 1:3.8 (g:ml), respectively. Additionally, the verification experiment revealed that the THA was (0.069 0±0.002 4) ℃, similar to the experiment value, which indicated that the model was reliable. The AFP separated and purified by column chromatography was electrophoresis level, the molecular weight was 6 400 u, and the THA was about (0.050 2±0.002 2) ℃.
