摘要
目的:探讨脉冲电场对肿瘤细胞黏附侵袭能力的影响及其可能的作用机制。方法:分别将场强为0 V/cm、500 V/cm、1 000 V/cm和1 500 V/cm的脉冲电场作用于宫颈癌Hela细胞,采用基质胶黏附实验观察脉冲电场对Hela细胞黏附能力的影响;采用Transwell小室实验观察脉冲电场对Hela细胞侵袭能力的影响;采用免疫荧光法检测脉冲电场作用后Hela细胞中MMP-2、TIMP-2的表达情况。结果:经脉冲电场作用后,Hela细胞黏附能力下降,侵袭能力受到抑制,MMP-2的表达水平下调,TIMP-2的表达水平上调。结论:脉冲电场能抑制Hela细胞的黏附和侵袭能力,这种抑制作用可能与其下调MMP-2的表达水平和上调TIMP-2的表达水平有关。
Objective: To explore the effects of pulse electric fields on adhesion and invasion of cervical cancer HeLa cells and the possible mechanism. Methods: Pulse electric fields with intensities of 0 V/cm, 500 V/cm, 1 000 V/cm, and 1 500 V/cm were used to treat cervical cancer HeLa cells, stromal adhesion experiment was used to observe the effects of pulse electric fields on adhesion of cervical cancer HeLa ceils; Transwell assay was used to observe the effects of pulse electric fields on invasion of cervical cancer HeLa cells ; immuno- fluorescence method was used to detect the expressions of matrix metalloproteinase-2' ( MMP-2 ) and tissue inhibitor of metalloproteinase 2 (TIMP-2) in cervical cancer HeLa ceils after treated by pulse electric fields. Results: After treated by pulse electric fields, adhesion ability of cervical cancer HeLa cells decreased, the invasion ability was inhibited, the expression level of MMP-2 was down-regulated and the expression level of TIMP-2 was up-regulated. Conclusion: Pulse electric fields can inhibit adhesion and invasion of cervical cancer HeLa cells, which may be correlated with down-regulation of MMP-2 and up-regulation of TIMP-2.
