氯化镉对大鼠原代星形胶质细胞凋亡的影响 被引量:4

摘要 目的研究氯化镉对星形胶质细胞凋亡的影响。方法取处于对数生长期的细胞,以终浓度为0(对照)、2.5、5、10、20、40μmol/L氯化镉溶液染毒12、24 h,采用MTT实验测定细胞的存活率;以终浓度为0(对照)、2.5、5、10、20μmol/L氯化镉溶液染毒12 h,采用Annexin V-FITC/PI双染流式细胞术测定细胞的凋亡率。结果与对照组相比,5~40μmol/L氯化镉染毒12、24 h时大鼠星形胶质细胞的存活率均较低,5~20μmol/L氯化镉染毒12 h时大鼠星形胶质细胞的凋亡率均较高,差异均有统计学意义(P〈0.05);且随着氯化镉染毒浓度的升高,大鼠星形胶质细胞的存活率呈下降趋势,而凋亡率呈上升趋势。结论氯化镉对星形胶质细胞具有明显的毒性作用,可诱导星形胶质细胞凋亡。 Objective To study the effects of cadmium chloride on apoptosis of rat primary astrocytes in cultured. Methods The astrocytes in logarithmic growth phase were exposed to cadmium chloride at the doses of 0 (control),2.5,5,10,20 and 40 μmol/L for 12 h and 24 h,then cell survival rate was measured by MTF assay;The astrocytes exposed to 0(control),2.5,5,10 and 20 μmol/L cadmium chloride for 12 h were used to detect apoptosis rate by flow cytometry stained with Annexin V-FITC/ PI. Results Compared with the control group, the astrocytes exposed to 5-40 μmol/L cadmium chloride for 12 h and 24 h revealed lower survival rate, the astrocytes exposed to 5-20 μmol/L cadmium chloride for 12 h revealed higher apoptosis rate with statistically significant difference (P〈0.05);And with the increase of cadmium chloride exposure level, the survival rate of the astrocytes decreased, the apoptosis rate increased. Conclusion Cadmium chloride may induce the apoptosis of astrocytes.
