响应面法优化甜叶菊毛状根3,5-二咖啡酰奎宁酸提取工艺

Optimization of Extraction Parameters of 3, 5-Dicaffeoylquinic Acid in Stevia rebaudianum Hairy Roots by Response Surface Methodology

摘要 以甜叶菊毛状根为材料,以3,5-二咖啡酰奎宁酸得率为考察指标,选取乙醇体积分数、液料比和超声提取时间3个因素,在单因素试验基础上,采用Box-Behnken设计3因素3水平的响应面试验方法,优化了超声辅助提取的主要工艺参数。结果表明,优化后的甜叶菊毛状根3,5-二咖啡酰奎宁酸提取参数为乙醇体积分数68%,液料比22∶1(m L/g),提取时间90 min;在优化条件下,该物质的得率为2.97%,与优化模型的预测得率3.01%基吻合,优化后的工艺参数可用于3,5-二咖啡酰奎宁酸的提取。 The extraction technology of 3, 5-dicaffeoylquinic acid in Stevia rebaudianum hairy roots by ulwasound-assisted exaction was studied. Based on the results of single factor expe6ments, three independent parameters named ethanol concentration, liquid to solid ratio and extraction time were optimized by response surface methodology (RSM). The results indicated that the optimal extraction parameters of 3, 5-dicaffeoylquinic acid were ethanol concentration 68%, liquid to solid ratio 22 : 1 (mL/g) and extraction time 90 min Under the optimal conditions, the extraction yield was 2.97%, which was in good consistent with the yield of the prediction model (3.01%). Consequently, the optimal extraction technology can be applied in the extraction of 3, 5 -dicaffeoylquinic acid.
