摘要
目的了解迷迭香提取物对果蝇的寿命和抗氧化能力的影响。方法以2d龄的Oregon K野生型雌性黑腹果蝇为模型,饲喂含有不同浓度(0、0.2、0.5、1.5 mg/ml)迷迭香提取物的培养基,每3d更换新的培养基并进行计数,计算果蝇平均寿命与最高寿命。测定饲喂45d果蝇体内抗氧化酶活力及抗氧化基因表达水平,同时进行百草枯与双氧水急性生存实验。结果给予迷迭香提取物后,果蝇的抗氧化能力明显增强,且呈剂量依赖关系。其中1.5mg/ml组果蝇的平均寿命及最高寿命较对照组显著延长(P<0.05),体内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活力及其m RNA表达水平显著提高(P<0.05),且丙二醛(MDA)的含量显著下降(P<0.05)。结论迷迭香提取物延长寿命与抗氧化作用可能是通过上调抗氧化基因的表达来实现的。
Objective To evaluate the effects of rosemary extract on lifespan and antioxidant capacity in female Drosophila melanogaster. Methods Female drosophila aged 2 days were treated with different doses(0, 0.2, 0.5, 1.5mg/ml) of rosemary extract. Fresh medium was replaced every 3 days and dead drosophila were counted until all drosophila died. Then, the maximum lifespan and mean lifespan were calculated. The antioxidant enzymes activities and the m RNA expression levels of antioxidant-related genes were measured after 45 days of feeding. Challenge test with paraquat or hydrogen peroxide was conducted. Results Compared with the control drosophila, the antioxidant capacity was increased after treatment with rosemary extract. The mean lifespan and maximum lifespan after exposed to 1.5 mg/ml rosemary extract were significantly prolonged(P〈0.05). The activity of superoxide dismutase(SOD) and catalase(CAT), as well as the m RNA expression of their encoding genes were also obviously increased(P〈0.05). The malondialdehyde(MDA) level was decreased significantly(P〈0.05). Conclusion The effects of rosemary extract on lifespan and antioxidant capacity are most likely via the up-regulation of antioxidant genes expression.
