摘要
目的利用鼠胚试验(mouse embryo assay,MEA)检测新建辅助生殖实验室培养系统是否适合胚胎培养。方法收集小鼠合子随机放入6个不同的培养箱中,观察各个箱中的囊胚形成情况;利用小鼠配子体外授精法将取出的卵子和精子体外授精,分别用COOK和Vitrolife两个系列的培养液处理配子、完成授精和胚胎培养,分别观察囊胚形成情况。结果共获得合子161枚,放入6个培养箱中的囊胚形成率分别为87.50%、100.00%、83.87%、95.00%、88.57%、92.11%。共获得51枚卵子,用COOK系列和Vitrolife的培养液授精后胚胎培养的囊胚形成率分别为87.50%和94.44%。结论利用收集小鼠合子和小鼠配子体外授精的胚胎培养技术对新建IVF实验室培养环境、培养系统及技术员水平评估,检测的结果符合各项质控标准。
Objective To detect whether embryo culture system of the newly established assisted reproductive laboratory is reliable for the human embryo culture by using mouse embryo assay(MEA). Methods Mouse zygotes with in vivo fertilization were cuhured in six different incubators, and mouse embryos were observed. Mouse embryos were cultured with in vitro fertilization (IVF), the gametes were cultured in COOK culture media and Vitrolife culture media, and blastulation were observed. Results A total of 161 zygotes were collected, and the blastocyst rate were 87.50 % ,100. 00 % ,83.87 % ,95.00 % ,88. 57 % ,92. 11% in six incubators respectively. A total of 51 ovums were obtained, and the blastocyst rate were 87.50 %, 94.44 % in COOK culture media and Vitrolife culture media respectively. Conclusion The results of application of embryos cultured by MEA to testing the culture environment,system and operating levels of the technicians in the newly built assisted reproductive laboratory are able to meet every quality control standards.
