摘要
[目的]研究棉花中SVP类基因的功能。[方法]通过同源克隆的方法在陆地棉中棉所36中克隆SVP的同源基因GhMADS29,对其进行Blast搜索比对和进化树分析以及荧光定量的表达模式分析。[结果]GhMADS29与猕猴桃的SVP4基因相似度最高,其cDNA序列含有9个外显子、8个内含子,不同时期顶芽的荧光定量结果表明它在花芽分化起始期的花芽中表达量最高,且不同组织的荧光定量分析表明它在叶片和顶芽中表达量较高。[结论]首次获得棉花SVP亚族基因,命名为GhMADS29,其GeneBank登录号为JQ682642。
[Objective] To study SVP gene from cotton.[Method]The SVP homologue gene Gh MADS29 was cloned from CCRI 36 upland cotton by homology cloning strategy.Blast alignment,phylogenetic tree and quantitative RT-PCR were conducted to analyze Gh MADS29.[Result]It was showed that Gh MADS29 was most similar to SVP4 from kiwifruit and that Gh MADS29 had nine exons and eight introns.Gh MADS29 expressed highest in 2SAM(shoot apical meristems at the second true leaf expanded stage) at which stage flower bud initiated differentiation.Expression analysis in different tissues showed that Gh MADS29 expressed very highly in leaves and apical buds.[Conclusion] Gh MADS29 is belong to SVP subfamily obtained for the first time in cotton and the accession number is JQ682642.
