摘要
目的应用荧光原位杂交技术(FISH)分析低形态学评分胚胎的染色体非整倍体情况并探讨低评分胚胎作为移植选择的安全性。方法收集IVF/ICSI周期中的D3低评分胚胎,卵裂球固定后,应用8种不同的染色体探针进行二轮FISH杂交检测细胞核。结果收集D3低评分胚胎40枚,获得178个卵裂球,成功固定核170个,固定成功率为95.5%(170/178);FISH杂交后163个核信号完整,杂交成功率为95.9%(163/170);40枚2PN胚胎中,未发现异常胚胎12枚,占30%,异常胚胎28枚,占70%。结论低评分胚胎染色体异常率高,临床应用于移植或冷冻时建议结合PGS。
[Objective]To analyze the chromosome aneuploidy of low grade embryos with fluorescence in situ hybridization(FISH) and to evaluate whether low grade embryo transplantation is a safe choice. [Methods]D3 low grade embryos were collected from IVF/ICSI cycles and fixed. Then the nuclei were detected using eight different chromosome probes in two rounds of FISH. [Results]Totally 178 blastomeres were obtained from 40 D3 low grade embryos in which 170 nuclei were fixed successfully. The fix rate was 95.5%(170/178). Complete FISH signals were obtained in 163 nuclei and the signal rate was 95.9%(163/170). In 40 2PN embryos, 12 were normal accounting for30% and 28 were abnormal accounting for 70%. [Conclusions]The aneuploidy rate of low grade embryos is high.When it is applied clinically to transplantation or cryopreservation, it is suggested to combine with PGS.
