摘要
目的探讨浸润性乳腺癌组织中14—3—3δ蛋白的表达与雌激素受体(ER)、孕激素受体(PR)的相关性。方法选择2012年12月~2014年10月重庆市公安消防总队医院手术切除的浸润性乳腺癌组织标本55例及其相应的癌旁正常组织31例为研究对象。分别将其作为浸润性乳腺癌组织组和正常乳腺组织组。采用免疫组化sP法检测乳腺组织中14—3—30r蛋白及ER、PR的表达情况;采用Pearson相关系数分析14—3—3δ蛋白表达水平与ER、PR的相关性。结果浸润性乳腺癌组织14—3—3δ蛋白及ER阳性表达率明显低于正常乳腺组织,差异均有统计学意义(P〈0.05或P〈0.叭);浸润性乳腺癌组织PR阳性表达率与正常乳腺组织比较,差异无统计学意义(P〉0.05);浸润性乳腺癌组织中,14—3—3δ盯蛋白阳性表达率明显低于ER、PR,差异均有统计学意义(均P〈0.05);正常乳腺组织中,14—3—3δ蛋白、ER及PR表达情况比较,差异无统计学意义(P〉0.05)。不同年龄、肿瘤大小、腋窝淋巴结转移情况的浸润性乳腺癌患者14—3—3δ蛋白阳性表达率比较,差异无统计学意义(P〉0.05);不同TNM分期、组织分型、分化程度、PR及ER表达情况的浸润性乳腺癌患者14—3—3δ蛋白阳性表达率比较,差异均有统计学意义(均P〈0.05)。Pearson相关性分析显示,14-3—3δ蛋白与ER阳性表达呈正相关(r=0.3334,P〈0.05),14—3—3δ蛋白与PR无相关性(P〉0.05)。结论14—3—3δ蛋白可能参与浸润性乳腺癌的发生、发展和转移,14—3—3δ与ER的联合检测可成为乳腺癌患者预后判断的重要指标。
Objective To explore the relationship between the expression of 14-3-3δ and ER, PR in invasive breast cancer tissue. Methods 55 cases of breast cancer tissues with surgical resection from December 2012 to October 2014 in Public Security Fire Corps Hospital of Chongqing City and 31 cases of normal tissue adjacent to carcinoma were selected as invasive breast cancer tissue group and normal breast tissue group respectively. The expression of 14-3-3δ protein, ER and PR in breast tissues were detected by immunohistochemical (SP method), and the relationship between the expression of 14-3-3δ and ER, PR were analyzed by Pearson correlation coefficient. Results The positive expres- sion rate of 14-3-3δ protein and ER in invasive breast cancer tissue was significantly lower than that of normal breast tissue respectively, with statistical difference (P 〈 0.05 or P 〈 0.01). The positive expression rate of PR in invasive breast cancer tissue was compared with normal breast tissue, with no statistical difference (P 〉 0.05). The positive ex- pression rate of 14-3-3δ protein in invasive breast cancer patients with different age, tumor size, axillary lymph node metastasis was compared respectively, with no statistical difference (P 〉 0.05). The positive expression rate of 14-3-3δ protein in invasive breast cancer patients with different TNM stage, histological type, differentiation degree, PR and ER was compared respectively, with statistical difference (P 〈 0.05). Pearson correlation analysis showed that 14-3-3δ protein was positively related with ER (r = 0.3334, P 〈 0.05), but there was no correlation between 14-3-3δ protein and PR (P 〉 0.05). Conclnsion The 14-3-3δ may be involved in occurrence, development and metastasis of invasive breast cancer, detection of 14-3-3δ combined with ER can be an important index for prognosis of patients with breast cancer.
