摘要
目的了解北京市丰台区污水中埃可病毒6型(echovirus 6,ECHO6)的基因特征。方法选择吴家村污水处理厂为监测点,2012年1~12月,每月采集监测点污水进行肠道病毒(enterovirus,EV)分离和鉴定,并对分离到的ECHO6进行VP1编码区核苷酸序列测定和分析。结果在吴家村污水处理厂监测点,共分离到64株病毒,其中ECHO6病毒12株,在非脊髓灰质炎(脊灰)肠道病毒(non-poliomyelitis entero-viruses,NPEV)分离株中比例最高。选择10株ECHO6进行VP1编码区核苷酸序列分析,结果表明10株病毒的VP1区核苷酸同源性为92.9%~100.0%,与D5基因型代表株的VP1区核苷酸同源性最高,为88.7%~94.2%。进化树分析结果显示,北京市丰台区ECHO6分离株与D5基因型代表株处于同一分支,并在D5进化分支的不同簇中。结论北京市丰台区污水中ECHO6分离株为D5基因型并在D5进化分支的不同簇中,并为丰台区人群肠道病毒的监测提供重要的补充信息。
Objective To study the genetic characteristics of echovirus 6in environmental sewage of Fengtai district in Beijing,China. Methods The Wujiacun Sewage Treatment Plant was chosen as the surveillance site,the sewage samples were collected monthly from January 2012 to December 2012.Viral isolation was performed from the sewage samples,and the VP1 coding region nucleotide sequence was obtained from all echovirus 6isolates. Results A total of 64 strains of enteroviruses were isolated from sewage water samples,including 12 strains of echovirus 6,which was the highest rate in none-polio enteroviruses.The whole VP1 coding region nucleotide sequence was amplified from 10 strains of echovirus 6.The homogeneity of VP1 nucleotide sequence ranges from 92.9% to 100.0%.When comparing with D5 subgenotype,the homogeneity is88.7%-94.2%.Phylogenetic tree analysis indicated that the Fengtai district echovirus 6strains belong to subgenotype D5. Conclusions The Fengtai district echovirus 6strains belong to subgenotype D5 with distinct clusters.Environmental surveillance provides important supplementary information for human enteroviruses infection.
