摘要 目的阐明转录因子Sox2在胃黏膜肠化生(intestinal metaplasia,IM)中的作用。方法 RT-PCR法检测人胃上皮细胞系GES-1,正常胃黏膜和肠黏膜活检组织中Sox2、Cdx2和Muc2 mRNA表达。设计合成Sox2 siRNAs并转染到GES-1细胞中使Sox2表达下调。RT-PCR检测对照组和Sox2干扰组肠上皮特异性基因Muc2和Cdx2 mRNA表达。结果 GES-1细胞、正常胃黏膜和肠黏膜活检组织中Sox2、Cdx2和Muc2 mRNA表达不同。特异性小干扰RNA明显下调GES-1细胞中Sox2 mRNA的表达。RT-PCR结果显示Sox2干扰组的Sox2 mRNA相对表达量(×10-3)低于对照组(70.7±6.9 vs 246.6±9.9,P=0.014),而Muc2 mRNA和Cdx2mRNA的相对表达量高于对照组(22.4±4.3 vs 0,P=0.012;646.2±204.2 vs 116.2±17.2,P=0.045)。结论 Sox2在胃黏膜IM的发生、发展中起着重要作用。 Objective To elucidate the effect of transcription factor Sox 2 in gastric intestinal metaplasia ( IM ) . Methods The expression of Sox2, Cdx2 and Muc2 mRNA in gastric epithelial cell line GES-1, biopsy tissues of normal gastric mucosa and intestinal mucosa were detected by RT-PCR.siRNAs targeted to Sox2 were designed , synthesized and transfected into GES-1 cells to down-regulate Sox2, then the expression of Muc2 and Cdx2 mRNA were detected by RT-PCR.Results Sox2, Cdx2 and Muc2 mRNA were variably expressed in GES-1, biopsy tissues of normal gastric mucosa and intestinal mucosa .Special siRNAs down-regulated the expression of Sox 2 mRNA significantly in GES-1 cells.RT-PCR showed that the relative expression ( ×10^-3 ) of Sox2 mRNA was lower in Sox2 interference group than that in control group (70.7 ±6.9 vs 246.6 ±9.9, P=0.014), while expression of Muc2 mRNA and Cdx2 mRNA were higher in Sox2 interference group than those in control group (22.4 ±4.3 vs 0, P=0.012;646.2 ±204.2 vs 116.2 ± 17.2, P=0.045).Conclusion Sox2 may play an important role in development of gastric IM .
