摘要
目的检测儿童特应性皮炎患者外周血白介素-17(IL-17)及白介素-8(IL-8)的表达水平,探讨其在儿童AD发病机制中的作用。方法采用双抗体夹心酶联免疫吸附试验(sandwich ELISA)检测60例儿童AD患者外周血IL-17、IL-8的表达情况,并用SCORAD评分标准判断AD患者病情严重程度。30例性别、年龄匹配的健康儿童作为对照组。结果 AD组患者血清IL-17的表达为(35.71±1.78)pg/ml,高于健康对照组(24.22±6.02)pg/ml,差异有统计学意义(t=3.66,P<0.05),且与SCORAD得分呈明显正相关(r=0.646,P<0.05);AD组患者血清IL-8的表达为(69.90±7.78)pg/ml,明显高于健康对照组(34.07±13.04)pg/ml,差异有统计学意义(t=4.72,P<0.01),且与SCORAD得分呈显著正相关(r=0.683,P<0.05)。结论儿童AD患者外周血IL-17、IL-8过度表达可能参与了AD的发病机理。
Objective To detect the expression level of interleukin-17(IL-17)and interleukin-8 (IL-8) in peripheral blood in the patients with atopic dermatitis of children, study the pathogenesis in the AD patients. Methods Peripheral blood was obtained from 60 patients with AD of children and 30 children as a control by sandwich enzyme-linked immunosorbent assay ( sandwich ELISA);with matched gender and age in both groups. And their relationship with AD disease severity were analyzed.AD disease severity scored with SCORAD criterion. Results The serum concentrations of IL-17 in the patients with AD of children (35.71 ±1.78pg/ml) was significantly higher than the healthy controls (24.22 ±6.02pg/ml) ( t =3.66,P 〈0.05) and positively correlated with SCORAD score in AD (r=0.646,P〈0.05).The serum concentrations of IL-8 in the patients with AD(69.90 ±7.78pg/ml)was significantly higher than the healthy controls(34.07 ±13.04pg/ml)(t=4.72, P〈0.01)and positively correlated with SCORAD score in AD (r=0.683,P〈0.05).Conclusions The high expression level of IL-17 and IL-8 in peripheral blood plays the important role in the pathogenesis of AD.
