mTOR和PTEN基因在大肠癌组织中的表达及临床意义 被引量:6

摘要 目的观察哺乳动物雷帕霉素靶蛋白(m TOR)和10号染色体同源丢失性磷酸酶基因和张力蛋白基因(PTEN)与大肠癌(CRC)的关系以及与CRC临床病理特征的相关性,为CRC的早期诊断和预后判断提供参考依据。方法外科手术中获取CRC组织及癌旁组织各72例,采用逆转录酶聚合酶链反应(RT-PCR)检测CRC组织及癌旁组织中m TOR和PTEN基因的RNA水平表达变化。结果 CRC组织中m TOR基因的表达明显高于癌旁组织,PTEN基因的表达明显低于癌旁组织,其差异均有统计学意义(P<0.05);CRC组织中m TOR、PTEN基因的RNA表达与性别、年龄、肿瘤部位差异无统计学意义(P>0.05),与不同肿瘤分化程度、有无淋巴结转移以及Dukes分期差异有统计学意义(P<0.05)。相关性分析显示,m TOR和PTEN在CRC中的表达呈负相关(P<0.05)。结论抑制m TOR基因的表达或者是增强PTEN基因的表达极有可能成为预防和或治疗CRC的有效方法之一。 [Objective] To observe the relationship between mTOR, PTEN and colorectal carcinoma (CRC), and the correlation between roTOR, PTEN and clinical and pathological features of CRC, provide the basis for the early diagnosis and prognosis judgement of CRC.[Methods]72 samples of CRC tissues and 72 samples of cancer adjacent tissues were collected by the surgery. The RNA level changes of roTOR and PTEN genes in CRC tissues and cancer adjacent tissues were detected by reverse transcriptase polymerase chain reaction technology (RT-PCR).[ Results]The expression level of mTOR in CRC tissues was higher than that in cancer adjacent tissues, while the expression level of PTEN in CRC tissues was lower than that in cancer adjacent tissues, and the differences were statistically significant (P〈0.05). The RNA expression levels of rnTOR and PTEN in CRC tissues were not related to the sex, age and tumor location (P〉0.05), but there were significant differences in the expression levels between different tumor differentiation degree, lymphatic metastasis, Dukes staging groups (P〈0.05). The correlation analysis shows that there was a negative correlation between roTOR expression and PTEN expression in CRC tissues (P〈0.05).[Conclusion]To inhibit mTOR expression or enhance PTEN expression could become an effective way to prevent and treat CRC.
