摘要
灵芝是我国传统大型药用真菌之一,属无隔担子菌亚纲,无褶菌目,灵芝科,灵芝属,常见的有赤芝[Ganoderma lucidum(Curtis:Fr.)P.karst]、紫芝[Ganoderma sinense(Zhao,Xu et Zhang)][1]。从赤芝[Ganoderma lucidum(Curtis:Fr.)P.karst]多糖肽中分离纯化具有活性成分GL-PPS,对其进行结构分析。经分离纯化得GL-PPS,通过高效液相凝胶渗透色谱法(high-performance gel-permeation chromatography,HPGPC)、气相色谱、甲基化分析和Smith降解等方法研究分析GL-PPS的结构。结果表明,GL-PPS的单糖组成为鼠李糖(Rha)、甘露糖(Man)和葡萄糖(Glc),物质的量比为14.9∶9.7∶75.4,其Mw为3.4×104,Mn为2.6×104;Mw/Mn为1.30。Mp为3.8×104。GL-PPS的主链由1,3-与1,4-两者交替连接的葡萄糖构成,在部分主链葡萄糖的6位有分支,非还原末端均由葡萄糖构成。
A water-soluble polysaccharide peptide designated GL-PPS,was isolated and purified from the heteropolysaccharide of Ganoderma lucidum fruit body. Using a range of techniques,such as high-performance gel-permeation chromatography,HPGPC gas chromatography,methylation analysis and Smith degradation methods,the sample was shown to be polysaccharide peptide,mainly composed of rhamnose,mannose and glucose with the amount of substance ratio 14. 9∶ 9. 7∶ 75. 4 and the weight-average molecular weight of 3. 4 × 104,number-average molecular weight of 2. 6 × 104. GL-PP was most branched of 1,3-and 1,4-alternate connection of glucose,whereas GL-PPS was mainly six branches glucose,a reduction at the end which was composed of glucose.
