摘要
为了研究云南不同地区烟草内生细菌的多样性及分布特征。在全省4个不同区域采集相同烟草品种33个样品,通过分离培养烟草内生细菌、16S r DNA序列分析获得不同烟区烟草内生细菌的多样性特征。从不同地区烟草叶片中共分离得到内生细菌372株,分别归属于11个属。以Bacillus属为优势菌群,已鉴定出7个种,包括B.pumilus,B.amyloliquefaciens,B.cereus,B.subtilis,B.flexus,B.simplex,B.megaterium;B.pumilus和Pantoea agglomerans类群细菌是4个地区共同的优势菌群;香农多样性指数由大到小依次为文山、大理、昭通、玉溪;其中玉溪和大理地区样品的相似性系数最高,为0.58,玉溪和昭通地区样品的相似性系统最低,为0.31。烟草地理分布对烟草内生细菌组成具有一定的影响,烟草内生细菌的种类多样性特征在不同地区间存在差异,部分内生细菌的分布存在地域专一性。研究结果为认识烟草内生细菌的特征及挖掘利用有益内生细菌奠定了基础,并为内生细菌在其它植物中的研究提供理论依据。
We researched the diversity and distribution characteristics for 33 sample of endophytic bacteria in Yunyan85 at 4 regions of Yunnan Province. These strains were isolated and identified into genera based on 16 S rRNA gene sequences. A total of 372 strains were isolated from the leaves of tobacco at different sampling sites and were identified into 11 genera. Bacillus were dominant bacteria in tobacco. 7 species,B. pumilus,B. amyloliquefaciens,B. cereus,B. subtilis,B. flexus,B. simplex,B. megaterium. B. pumilus and Pantoea agglomerans were dominant bacteria in 4 regions. The Shanno Index from high to low was Wenshan,Dali,Zhaotong,Yuxi. The similarity coefficient of Yuxi and Zhaotong were the lowest( 0. 31). The similarity coefficient of Yuxi and Dali were the highest( 0. 58). Similarity comparison of endophytic bacteria community compositions in different regions suggested geographical distribution had a certain impact on the community composition of endophytic bacteria. The diversity and distribution of the endophytic bacteria varied from different regions,which indicated that the distribution of endophytic bacteria had geographic specificity. This studies could offer foundation for screening of beneficial endophytic bacteria and provide fundamental theory for exploiting and using it from other plant.
