纳豆芽孢杆菌发酵制备高蛋白微生态血粉的研究 被引量:4

摘要 本试验以水解度作为血粉发酵的评价指标,利用Box-Behnken试验设计和响应面分析法进行回归分析,优化纳豆芽孢杆菌发酵血粉的工艺参数。结果表明,发酵温度和转速与血粉水解度存在显著相关性,优化后的最佳发酵条件为发酵温度36.6℃、接种量6.6%、转速202 r/min。在该条件下血粉水解度达到29.1%,发酵液中可溶性氮和游离氨基酸含量分别增加至4.24 g/L和1.09 g/L,蛋白酶活力达到1576 U/m L,纳豆芽孢杆菌活菌数量达到8.8×108cfu/m L。 In this study,Box-Behnken design and response surface analysis were applied to optimizing condition for blood powder fermentation by Bacillus natto,which considered hydrolysis degree as evaluation indexes.Experimental resuits showed that temperature and rotation speed all had significant influence on hydrolysis degree.The optimal condition for the maximal hydrolysis degree of the blood powder were temperature 36.6℃,inoculum concentration 6.6% and rotation speed 202 r/min.Under this optimal condition,the maximum hydrolysis degree of the fermented blood powder was 29.1%, and the soluble nitrogen and free amino acid content in the fermentation broth increased to 4.24 g/L and 1.09 g/L,respectively.Meanwhile,protease activity and total viable Bacillus natto amount in the fermentation broth reached to 1576 U/mL and 8.8×10^8 cfu/mL,respectively.
