海洛因成瘾者全脑功能网络拓扑结构异常的静息态功能磁共振成像研究 被引量:2

摘要 目的探讨海洛因成瘾者全脑功能网络的拓扑结构是否异常。方法 17名海洛因成瘾者(HDIs)及15名年龄、性别与之匹配的健康对照者(NCs)参加本研究。采用1.5T磁共振扫描仪进行静息态f MRI数据采集,使用图论法构建HDIs及NCs的脑功能网络,采用两独立样本t检验的统计学方法比较两者功能网络拓扑属性的组间差异。结果与NCs相比,HDIs具有更小的归一化聚类系数(P=0.049)及更低的小世界性(P=0.035)。HDIs认知控制网络区域的主要节点向心性显著下降,包括两侧中央扣带回、左额中回及右楔前叶,而左侧海马的主要节点向心性显著增加。左海马的节点向心性与海洛因成瘾的持续时间呈正相关。结论 HDIs的全脑功能网络遭到破坏,这有助于阐释海洛因成瘾的机制。 Objective To investigate whether heroin addiction changes the topological organization of whole-brain functional networks. Methods Seventeen heroin-dependent individuals (HDIs) and 15 age, gender matched normal controls (NCs) were enrolled, and the resting-state functional magnetic resonance images (RS-fMRI) were acquired from these subjects using a 1.5 T MR scanner. We constructed the brain functional networks of HDIs and NCs, and compared the group differences in network topological properties using graph theory method. Results We found that the HDIs showed decreases in the normalized clustering coefficient and in small-worldness compared to the NCs. Furthermore, the HDIs exhibited significantly decreased nodal centralities primarily in regions of cognitive control network, including the bilateral middle cingulate gyrus, left middle frontal gyms, and right precuneus, but significantly increased nodal centralities primarily in the left hippocampus. Moreover, nodal centralities in the left hippocampus were positively correlated with the duration of heroin addiction. Conclusion Our results indicated that disruptions occur in the whole-brain functional networks of HDIs, findings which may be helpful in further understanding the mechanisms underlying heroin addiction.
