冰片配伍灯盏花素对大鼠脑缺血再灌注损伤血脑屏障通透性的影响 被引量:7

Effects of borneol combined with breviscapine on blood-brain barrier permeability after ischemia reperfusion injury in rats

摘要 目的:研究冰片配伍灯盏花素后对脑缺血再灌注大鼠血脑屏障通透性的影响。方法:50只雄性大鼠随机分为假手术组、模型组、冰片组、灯盏花素组及冰片配伍灯盏花素组。线栓法建立大鼠脑缺血再灌注模型,缺血2 h,再灌注24 h进行检测。伊文思蓝法检测血脑屏障通透性,免疫印迹法检测ZO-1和Claudin-5蛋白的表达,RT-PCR法检测ZO-1和Claudin-5基因的表达。结果:冰片配伍灯盏花素能显著降低脑组织中EB浓度(9.87±1.15),提高ZO-1和Claudin-5蛋白(0.83±0.06,1.05±0.07)和mRNA(1.51±0.07,1.58±0.08)的表达。结论:冰片配伍灯盏花素对血脑屏障的保护作用可能与冰片促进灯盏花素透过血脑屏障有关。 Objective: To investigate the effects of borneol combined with breviscapine on blood-brain barrier permeability after ischemia reperfusion injury in rats. Methods: 50 male rats were randomly divided into the sham operation group,the model group,the borneol group,the breviscapine group and borneol combined with breviscapine group. The rat ischemia reperfusion model was established by MCAO. The BBB permeability was detected by Evans Blue( EB) leakage,and the protein and mRNA expression of ZO-1 and Claudin-5 was detected by WB and RTPCR. Results: The borneol combined with breviscapine group could decrease the permeability of BBB and increase the expression of ZO-1and Claudin-5 protein( 0. 83 ± 0. 06,1. 05 ± 0. 07) and mRNA( 1. 51 ± 0. 07,1. 58 ± 0. 08). Conclusion: The protective effects of borneol combined with breviscapine on BBB may relate with that borneol promote breviscapine permeating the BBB.
