摘要
目的:研究石菖蒲挥发油的抗惊厥作用及其机制。方法:采用四种动物惊厥模型,测定石菖蒲挥发油抗惊厥效应并分析时效、量效关系及作用机制。结果:石菖蒲挥发油200、300、400 mg/kg灌胃,能显著提高电刺激大鼠皮层惊厥阈值,其中400mg/kg组药后1h、2h、4h惊厥阈值576.8μA、615.9μA、622.0μA明显高于石菖蒲其它剂量组惊厥阈值536.3μA、561.7μA、562.9μA和549.8μA、586.3μA、584.9μA;而丙戊酸镁90 mg/kg灌胃作用虽强,但其维持时间短;石菖蒲挥发油各剂量组对最大电休克惊厥实验(MES)、戊四唑惊厥实验(MET)抗惊厥效应明显优于生理盐水对照组,且均具有剂量依赖性对抗作用;海人酸模型对照组大鼠PKC阳性细胞数明显高于假手术对照组,石菖蒲挥发油300、400 mg/kg灌胃组PKC表达68.3、45.0明显低于模型对照组90.4,且呈剂量相关性。结论:石菖蒲挥发油对大、小鼠多种惊厥模型有效,是一种作用持续时间较长,抗痫谱广的抗癫痫药物,调控PKC表达,进而减少神经细胞凋亡可能是其抗癫痫作用机制之一。
Objective: To study the anticonvulsants and its mechanism of the volatile oil of Acorus tatarinowii Schott. Methods: The anticonvulsive effect and its mechanism of Acorus tatarinowii Schott were determined by four animal convulsion models and the time relationship and the concentration-response relationship were determined,too. Results: volatile oil of Acorus tatarinowii Schott 200,300,400 mg / kg ig,could significantly improve the rat cortex convulsions threshold by the electrical stimulation( P 〈0. 05),and the convulsions threshold576. 8μA,615.9μA,622. 0μA of the group of 400 mg / kg was significantly higher than other groups at the time 1h,2h,4h after medicine given 536. 3μA,561. 7μA,562. 9μA and 549. 8μA,586. 3μA,584. 9μA( P 〈0. 05); VPM90 mg / kg,ig,the effect is stronger,but its work time was shorter;the anticonvulsive effects of all the groups of the volatile oil of Acorus tatarinowii Schott were obviously better than that of normal saline control group for MES and MET( P 〈0. 05) and the volatile oil of Acorus tatarinowii Schott had a dose-dependent combat role; PKC had a higherlevel expression and increased significantly after KA given( P 〈0. 01) and the volatile oil of Acorus tatarinowii Schott could significantly reduce the expression of PKC,that was,68. 3、45. 0,inferior significantly to that of model group 90. 4 and the higher expression after the higher the volatile oil of Acorus tatarinowii Schott group( P 〈0. 01). Conclusion: the volatile oil of Acorus tatarinowii Schott was effective in the multiple seizures in mice and rats model,and was kind of drug,whose function duration was longer,and whose resistant spectrum of epilepsy was wider. regulating the expression of PKC,and thus reducing the nerve cell apoptosis may be one mechanism of its antiepileptic effect.
