摘要
目的:研究温阳化瘀方对2型糖尿病模型大鼠空腹血糖、抵抗素、脂联素、胰岛素和胰岛素抵抗指数的影响。方法:除正常对照组外,各组大鼠禁食12h,按60 mg/kg剂量尾静脉一次性快速注射链佐菌素制备动物模型,并以高脂高糖饲料喂养,饮用蒸馏水,连续喂养12周。于模型制作当日各给药组大鼠分别灌胃给予药物治疗,每日一次,连续12周,其中降脂组给予阿托伐他汀1.25 mg/kg,降糖组给予盐酸二甲双胍670 mg/kg,试药四个剂量组分别给予温阳化瘀方浓缩液4.5、6、12、18 g/kg,正常对照组、模型组给予同样量的生理盐水。分别于第4周、第8周、第12周尾静脉取血测量血糖值。采用酶联免疫吸附分析方法测定血清抵抗素、脂联素、胰岛素水平,计算胰岛素抵抗指数。结果:温阳化瘀方有降血糖作用,6g/kg开始起效,18g/kg剂量组12周时血糖值为8.07±0.39mmol/L(正常对照组8.73±0.37mmol/L)。试验药物12、18g/kg剂量组均能降低2型糖尿病模型大鼠血清抵抗素、脂联素、胰岛素含量,与正常对照组和模型组比较,具有非常显著性差异,6g/kg剂量组也有显著的差异。温阳化瘀方18g/kg剂量组作用较强,12周时呈显著变化,血清抵抗素0.12±0.03(正常对照组0.12±0.02μg/L);血清脂联素0.29±0.07μg/L(正常对照组0.24±0.03μg/L);胰岛素0.66±0.04m U/L(正常对照组0.68±0.11m U/L),并对胰岛素抵抗指数有一定影响。结论:温阳化瘀方对2型糖尿病模型大鼠血糖、血清抵抗素、脂联素、胰岛素水平及胰岛素抵抗均有调节作用,提示温阳化瘀方有调节脂肪和糖代谢的作用,其作用与剂量呈正比例关系。
Objective: To explore the effects of decoction of Wen Yang Hua Yu Fang on metabolic syndrome. Methods: 70 male specified-pathogens free( SPF) Wistar rats,were divided into seven groups in randomized manner: control with distilled water diet; high fat and( 49% fat) and high sugar( 10% edible sugar)( model group); high fat and sugar + treated with lipitor( lipid-lowering group); high fat and sugar + treated with metformin( hypoglycemic group); high fat and sugar + treated with Wen Yang Hua Yu Fang decoction 4. 5g / kg,6g / kg,12 g / kg,18 g / kg dose groups. The rats of every group were taken in high fat and high sugar and taken in addition to the normal control group,a group rats fasting 12 h,intravenous single rapid injection preparation of animal model of streptozotocin 60 mg / kg by except control group. In addition to the normal control group,other groups of rats were feeding with high fat and high sugar. In addition to the normal control group,other groups and model at all for set of rats given by ig drug treatment,once a day,for 12 weeks. Among them,the rats in lipid-lowering group were taken 1. 25 mg / kg lipitor; hypoglycemic group rats 670 mg / kg / d metformin; Wen Yang Hua Yu Fang decoction 4. 5g / kg,6g / kg,12 g /kg,18 g / kg dose group rats 4. 5g / kg,6 g / kg,12 g / kg and 18 g / kg Wen Yang Hua Yu Fang decoction concentrated solution differently. Control group and model group rats were taken normal salt comadministration by gavage by equal content. The blood glucose of all groups were measured with blood the tail vein of rats,And the level of serum resistin,adiponectin and insulin were detected by enzyme-linked immunosorbent assay( ELISA). And insulin resistance( HOMA-IR) index was counted. Results: The blood glucose was decreased while Wen Yang Hua Yu decoction was taken. The level of resistin adiponectin and insulin was of a very significant difference after the decoction were taken for12 weeks while compared with control group and model group( all P〈0. 01). The level of insulin and HOMA-IR index were not significant difference compared with the control group,lipid-lowering group and hypoglycemic group( P〉0. 05),but there was a significant difference compared with the model group( all P〈0. 01). The effective dose was from 6g / kg to 18 g / kg. It did'nt have effective at 4. 5g / kg. Conclusion: The Wen Yang Hua Yu Fang decoction can regulate metabolic syndrome model rats blood glucose,resistin,adiponectin,insulin and HOMA-IR index. It was suggested that the decoction of Wen Yang Hua Yu Fang be of effect on regulation fat and saccharide metabolism. The effective the decoction is relative to the dose.
