摘要
目的应用PCR-DGGE技术评价西芹对昆明小鼠肠道菌群平衡的影响。方法给予小鼠高脂饲料,复制高脂模型,模型复制成功后,每日给予小鼠西芹提取液灌胃,连续28 d。分别在实验的0、14和42 d,收集每只小鼠粪便,提取基因组DNA,应用PCR-DGGE(聚合酶链式反应——变性梯度凝胶电泳)技术获得肠道菌群分子指纹图谱,针对相似性、多样性分析及优势条带的序列进行分析。结果高脂饲料喂养后肠道益生菌约氏乳杆菌等趋近消失,出现了特异菌真杆菌,而西芹灌胃组和食用基础饲料组真杆菌数量显著减少,且西芹灌胃组作用大于食用基础饲料自然恢复组。结论西芹对高脂小鼠肠道菌群的平衡有显著的恢复作用。
Objective To study the effect of Celery (Apium graveolens ) on the intestinal flora in hyperlipedimia mouse models. Methods Kunming mice were fed in two groups, Control ( normal diet) and High Fat diet (HFD) groups, for 42 days. The HFD group was divided into two subgroups, Normal diet (ND) and Celery diet (CD) , after 28 days on high fat diet. The CD group received oral gavage of celery extracts for 28 days. PCR-DGGE tech- nology and sequence analysis was applied to obtain the intestinal flora molecular fingerprint, and the similarity and diversity of DNA extracted from stool were analyzed at days 0, 14 and 42 of experiment. Results Probiotic bacteria Lactobacillus johnsonii nearly disappeared and Eubacteria were detected in HFD group. After Celery gavage, the number of Eubacteria in CD group reduced significantly, to an extent greater than in ND group. Conclusion Cel- ery has a recovery effect on the balance of intestinal flora of mice fed with high fat diet.
