鼻咽癌组织中DAPK基因启动子甲基化的研究

摘要 目的:分析鼻咽癌组织中DAPK基因启动子的甲基化状态,探讨DAPK基因启动子甲基化与鼻咽癌的关系。方法:应用甲基化特异性PCR技术检测48例鼻咽癌组织、26例慢性鼻咽黏膜炎症组织的DAPK基因启动子甲基化状态,比较鼻咽癌和慢性鼻咽黏膜炎症组织的DAPK基因启动子甲基化率。结果:鼻咽癌组织的DAPK基因启动子甲基化率为75%,而慢性鼻咽黏膜炎症组织中未检测到DAPK基因启动子甲基化。结论:鼻咽癌组织中DAPK基因启动子存在高甲基化水平,检测DAPK基因启动子甲基化或许能为鼻咽癌的诊断提供依据。 Objective:To analyze methylation of death - associated protein kinase gene(DAPK)promoter in na-sopharyngeal carcinoma,and to explore the relationship between carcinogenesis of nasopharyngeal carcinoma and methylation of DAPK gene promoter. Methods:Methylation specific - polymerase chain reaction assay was designed to detect methylation status of DAPK gene promoter from 48 patients with nasopharyngeal carcinoma and 26 patients with chronic rhinitis,the rate of methylation was compared for patients with nasopharyngeal carcinoma and chronic rhinitis. Results:The frequency of promoter methylation of DAPK gene was 75% in nasopharyngeal carcinoma biopsy sam-ples,while methylation was not found in chronic rhinitis biopsy samples. Conclusion:Aberrant promoter methylation of DAPK gene is found in nasopharyngeal carcinoma,and assay of methylation of DAPK gene promote may be helpful for diagnosis of nasopharyngeal carcinoma.
