摘要
目的:探讨花椒多酚类化合物总提取物(ZPPC)对卵巢摘除模拟更年期的雌性ICR模型小鼠的抗抑郁作用及可能机制。方法:采用经典的行为绝望抗抑郁模型,即小鼠悬尾实验和强迫游泳实验。观察ZPPC对应激小鼠绝望不动时间的影响;同时观察ZPPC对小鼠自主活动的影响。采用荧光分光光度法测定ZPPC对小鼠脑内单胺氧化酶活性的影响。结果:与OVX组相比较,ZPPC(50、100、200 mg/kg)明显缩短小鼠悬尾和强迫游泳实验中的不动时间,其作用类似于阳性对照药氟西汀(10 mg/kg);ZPPC还明显抑制脑内单胺氧化酶的活性。结论:ZPPC预给药在更年期小鼠行为绝望模型中显示出抗抑郁作用,其机制可能与抑制脑内单胺氧化酶活性有关。
Objective:To explore the antidepressant effect of chronic pre-administration with procyanidins extract from Zanthoxylum bungeanum Maxim (ZPPC) on ovariectomized model mice and its possible mecha-nism. Methods:The animals behavioral despair models of depression, including the tail suspension and forced swimming tests, were used to explore the antidepressant effects of ZPPC. The locomotor activity test was used to detect the change of locomotor activity. Meanwhile, the monoamine oxidase activity in the mouse brain was also determined. Results:The results showed that ZPPC (50, 100, 200 mg/kg) signiifcantly reduced the duration of immobility in both tail suspension and forced swimming tests. The effective doses did not alter locomotot activity, resembling the selectivity of the positive control drug lfuoxetine (10 mg/kg) in relieving depression. Moreover, ZPPC was found to inhibit monoamine oxidase activity in the mouse brain. Conclusion:These ifndings suggest that ZPPC exerts antidepressant effect in animals behavioral despair tests and the underlying mechanism may in-volve the central monoaminergic systems.
