心脏直接重编程技术--由非心肌细胞诱导的心肌细胞参与心脏修复

摘要 以心肌细胞损伤以及心功能受损为特征的心脏病是全球发病和死亡的首要原因。成人心肌细胞再生能力有限,替代损伤心肌的再生疗法能够恢复心脏功能,每年有数百万患者因此受益。直接重编程的最新进展为心脏再生提供了有力的工具。心肌成纤维细胞直接重编程可规避干细胞疗法在临床应用中产生的许多问题。本综述将讨论心脏重编程领域的方法、临床应用进展及挑战。 As the leading cause of morbidity and mortality worldwide, heart disease is characterized as the signifi-cant loss of cardiomyocytes and impaired cardiac function due to the limited regenerative capacity of adult myocardi- um. Regenerative therapies that replace damaged myocardium could restore the cardiac function and benefit millions of patients annually. Recent advances in direct reprogramming have provided powerful tools for heart regeneration. Direct reprogramming of the resident cardiac fibroblasts into cardiomyocytes can circumvent many of the issues that stem cell-based therapies have in clinical application. In this review we will discuss the progress, approaches, clini- cal application and challenges in the field of cardiac reprogramming.
