摘要
以植株茎段为外植体,进行了甜樱桃矮化砧木品种‘吉塞拉6号'离体快速繁殖技术研究。结果显示:外植体培养于改良MS+1 mg/L ZT+0.5 mg/LBA+0.2 mg/L NAA+0.5 g/L CH,30 d后腋芽萌发率达81.3%;丛芽增殖的适宜培养基是:改良MS+0.5 mg/L BA+0.5 mg/L KT+0.1 mg/L NAA+0.01 mg/L TDZ,增殖系数达5.8;1/2MS+1.0 mg/L IBA+0.2 mg/L NAA是诱导甜樱桃试管苗生根的适宜培养基,生根率达98.3%;适宜试管苗移栽的基质配比为泥炭:椰糠:珍珠岩=1:1:1(v:v:v),试管苗移栽成活率达98%以上。
The stem segments were used as explants,and rapid propagation of gean dwarf stock variety‘Gisela 6’were studied.The results showed that the explants were cultured with the modified MS +1 mg/L ZT +0.5 mg/L BA +0.2 mg/L NAA +0.5 g/L CH,and 30 days later the germination rate of axillary buds was 81.3%;The suitable medium for the bud multiplication was the modified MS +0.5 mg/L BA +0.5 mg/L KT +0.1 mg/L NAA +0.01 mg/L TDZ,and the propagation coefficient reached 5.8;The suitable medium of inducing the test-tube plantlets to root was 1 ?2 MS +1.0 mg/L IBA +0.2 mg/L NAA,and the rooting percentage was up to 98.3%;For the test-tube plantlets to be transplanted the suitable volumetric ratio of the substrate peat +coconut husk +perlite was 1∶1∶1(v-v?v),and the survival rate of test-tube plantlets was over 98%.
