甜樱桃砧木品种‘吉塞拉6号’的离体培养与快速繁殖

In vitro culture and rapid propagation of gean rootstock 'Gisela 6'

以植株茎段为外植体,进行了甜樱桃矮化砧木品种‘吉塞拉6号'离体快速繁殖技术研究。结果显示:外植体培养于改良MS+1 mg/L ZT+0.5 mg/LBA+0.2 mg/L NAA+0.5 g/L CH,30 d后腋芽萌发率达81.3%;丛芽增殖的适宜培养基是:改良MS+0.5 mg/L BA+0.5 mg/L KT+0.1 mg/L NAA+0.01 mg/L TDZ,增殖系数达5.8;1/2MS+1.0 mg/L IBA+0.2 mg/L NAA是诱导甜樱桃试管苗生根的适宜培养基,生根率达98.3%;适宜试管苗移栽的基质配比为泥炭:椰糠:珍珠岩=1:1:1(v:v:v),试管苗移栽成活率达98%以上。

The stem segments were used as explants,and rapid propagation of gean dwarf stock variety‘Gisela 6’were studied.The results showed that the explants were cultured with the modified MS ＋1 mg/L ZT ＋0.5 mg/L BA ＋0.2 mg/L NAA ＋0.5 g/L CH,and 30 days later the germination rate of axillary buds was 81.3%;The suitable medium for the bud multiplication was the modified MS ＋0.5 mg/L BA ＋0.5 mg/L KT ＋0.1 mg/L NAA ＋0.01 mg/L TDZ,and the propagation coefficient reached 5.8;The suitable medium of inducing the test-tube plantlets to root was 1 ？2 MS ＋1.0 mg/L IBA ＋0.2 mg/L NAA,and the rooting percentage was up to 98.3%;For the test-tube plantlets to be transplanted the suitable volumetric ratio of the substrate peat ＋coconut husk ＋perlite was 1∶1∶1（v-v？v）,and the survival rate of test-tube plantlets was over 98%.
