艾灸各因素对APP/PS-1双转基因AD小鼠脑中DNA、蛋白质、脂质氧化应激的影响 被引量:31

Effects of different factors of moxibustion on DNA, protein and lipid oxidative stress in brain issue of APP/PS-1 transgenic AD mice

摘要 目的:通过对比艾灸各因素对APP/PS-1双转基因阿茨海默病(AD)小鼠大脑皮质中DNA、脂质和蛋白质氧化损伤产物的影响,探讨艾灸的抗衰老机制。方法:将5月龄C57BL/6小鼠作为空白对照组(6只),同龄APP/PS-1小鼠随机分为模型组、艾灸组、无烟灸组、艾烟组(6只/组)。艾灸与无烟灸组灸治小鼠关元穴。艾烟组小鼠束缚暴露于5-15mg/m3烟气环境,空白对照组与模型组小鼠束缚暴露于正常环境,20min/d,6d/周,共8周。于第9周取材,ELISA法检测大脑皮质中8-羟基脱氧鸟苷(8-Ohd G)、4-羟基壬烯醛(4-HNE)、3-硝化酪氨酸(3-NT)含量。结果:皮质中8-Ohd G、4-HNE、3-NT含量,模型组高于空白对照组(P<0.05);艾灸组、艾烟组、无烟灸组显著低于模型组(P<0.01)。结论:艾灸各因素可通过影响APP/PS-1小鼠脑内8-Ohd G、4-HNE、3-NT含量,改善脑内氧化应激状态,起到延缓脑组织衰老作用。 Objective: By comparing the effects of different factors of moxibustion on DNA, lipid and protein oxidized products in the cerebral cortex of APP/PS-1 AD mice, to investigate the potential anti-aging mechanism of moxibustion. Methods: Five-month old C57BL/6 mice (n=6) were used for normal control group. Same age APP/PS-1 mice were randomly divided into four groups (n=6): model group, moxibustion group, smokeless moxibustion group, and moxa smoke group. Moxibustion was performed with a moxa stick on Guanyuan (RN4) for 20min per day, 6 days per week for a total of 8 weeks. In the smokeless moxibustion, the procedure was same except that moxa stick was exchanged by smokeless one. Mice of the moxa smoke group were exposed to smoke, whose concentration was from 5 to 15mg/m^3, however, the normal control mice were just exposed to normal environment. The mice were sacrificed and the tissues were harvested for ELISA test in the 9th week. Results: The level of 8-OHdG, 4-HNE, and 3-NT in the model group was higher than that in the normal control group (P〈0.05); moxibustion group, smokeless moxibustion group, and moxa smoke group was lower than model group (P〈0.01). Conclusion: Moxibustion, smokeless moxibustion, and moxa smoke may change the oxidative stress of brain via inhibiting the production of 8-OHdG, 4-HNE, 3-NT in cortex neurons, and delaying the brain aging.
