通腑泻肺方对脓毒症大鼠肺p38活性的调控作用(英文) 被引量:2

Effects of Tongfuxiefei Formula on regulation of p38 activation in the lung of rats with sepsis

摘要 目的:探讨通腑泻肺方对脓毒症大鼠肺p38活性的调控作用。方法:将32只大鼠随机分为正常组、假手术组、模型组、治疗组,每组8只。采用盲肠结扎穿孔术制备脓毒症模型,治疗组予以通腑泻肺方灌胃。应用透射电子显微镜观察肺组织的病理改变;采用Western blotting法检测p38和p-p38的表达,用RT-PCR法分析p38m RNA的表达。结果:电镜下,模型组肺组织炎性反应明显,而治疗组肺组织炎性反应不明显。与正常组及假手术组相比,模型组p38表达水平明显升高(P<0.01),p-p38表达水平明显升高(P<0.01),p38 m RNA表达水平明显升高(P<0.01)。与模型组相比,治疗组p38表达水平降低(P<0.05),p-p38表达水平明显降低(P<0.01),p38 m RNA表达水平降低(P<0.05)。结论:通腑泻肺方对脓毒症大鼠肺组织有明显改善作用,其机制可能与其具有调控p38活性,减轻肺组织损伤有关。 Objective: To investigate the role of Tongfuxiefei Formula in the regulation of p38 activation in lungs of rats with sepsis. Methods: In this study, 32 rats were randomly assigned to one of the following four groups: the control group, the sham-operated group, the model group, and the treatment group. Each group comprised 8 rats. Sepsis was induced by cecum ligation and puncture, and medicine was administered via the intragastric route. Rats in the treatment group were administered Tongfuxiefei Formula. Scanning transmission electron microscopy was used to examine lung tissue for pathological changes. Protein expression of p38 and p-p38 was ascertained by Western blotting analysis. Reverse transcription polymerase chain reaction (RT-PCR) was used to analyze p38 mRNA gene expression. Results: Compared with the model group, there was no obviously inflammatory response in the treatment group under electron microscopy. A significant increase was observed in the expression levels of p38, p-p3g, and p38 mRNA in the model group comparing with the blank control and sham-operated groups (P〈0.01). The expression levels of p38, p-p38, and p38 mRNA decreased significantly in the treatment group (P〈0.05, P〈0.01, and P〈0.05, respectively) comparing with the model group. Conclusion: Tongfuxiefei Formula significantly protects against tissue damage in the lungs of rats with sepsis. The improvement observed in the lung tissue may be due to the ability of Tongfuxiefei to regulate p38 activation, thereby minimizing lung tissue damage.
