摘要
目的:应用电子顺磁共振(ERS)技术评价芪味理肺汤对支气管哮喘模型大鼠血清清除氧自由基能力的影响。方法:将SPF级SD雄性大鼠60只随机分为正常组、模型组、西药组和芪味高、中、低剂量组。除正常组外均建立哮喘模型,治疗后肺组织病理染色、支气管细胞灌洗液(BALF)细胞计数和肺功能指标评价芪味理肺汤疗效,ESR技术评价血清清除氧自由基能力。结果:与正常组比较,模型组肺组织炎性细胞浸润等病理表现明显,治疗后芪味高剂量组、西药组肺组织病理改善优于其他治疗组。与正常组相比,模型组BALF涂片嗜酸性粒细胞(EOS)计数显著升高(P<0.01),与模型组相比,中药组可显著减少EOS数量(P<0.01)。血清清除氧自由基柱状图及ESR波谱图中可见中药组清除氧自由基的能力较强,其中芪味中、高剂量组清除能力优于西药组,但组间比较差异无统计学意义。结论:芪味理肺汤中、高剂量组可改善血清清除氧自由基能力,可能通过抗氧化机制起到治疗哮喘的作用。
Objective: To evaluate the effect of Qiwei Lifei Decoction on oxygen free radical scavenging capacity of bronchial asthma rats serum with Electron Spin Resonance Spectroscopy (ERS). Methods: Sixty SD rats were randomly divided into six groups: normal group, model group, western medicine group, Qiwei high, middle and low dose groups. The bronchial asthma rats model was established except for the normal group. The pulmonary function (PF), pulmonary histopathology and bronchoalveolar lavage fluid (BELF) cells count index were detected. The ability of serum to remove oxygen free radical was evaluated by ESR. Results: Compared with normal group, lung pathological was increased significantly in model group. Compared with other treatment groups, lung tissue pathology was reduced significantly in Qiwei high dose group and western medicine group. Compared with normal group, BALF smear counting EOS was increased significantly in model group (P〈0.01). Compared with model group, Chinese medicine could significantly decrease the number of EOS (P〈0.01). From serum scavenging oxygen free radicals histogram and ESR spectrum diagram, traditional Chinese medicine group had higher ability to remove oxygen free radicals, Qiwei high dose group and middle dose group were superior to western medicine group, with no statistically significant difference among the three groups. Conclusion: Qiwei high and middle dose group could improve the ability of serum to remove oxygen free radical, which may play a role in the treatment of asthma through the antioxidant mechanism.
