摘要
目的:观察"通督启神"电针疗法对老年性痴呆(AD)模型小鼠空间学习记忆能力与大脑神经元葡萄糖代谢水平的影响,评价"通督启神"电针法治疗AD的疗效。方法:将30只SAMP8小鼠随机分为老年痴呆组、药物对照组和"通督启神"电针组,10只同背景SAMR1小鼠作为正常对照组。"通督启神"电针组,穴取"百会"和"印堂"接电针,电针频率为2Hz,留针20min/d,并于电针治疗前点刺"水沟"穴;药物对照组,取多奈哌齐,按0.65μg·g-1·d-1,灌胃给药;正常对照组与老年痴呆组仅作束缚。治疗15d后,运用Morris水迷宫实验观察各组小鼠行为学变化,小动物正电子发射断层扫描技术观察各组小鼠大脑葡萄糖代谢水平的变化。结果:与老年痴呆组相比较,"通督启神"电针组小鼠的空间学习记忆能力、大脑神经元每克组织葡萄糖的摄取率均有显著提高(P<0.05);与药物对照组相比较,"通督启神"电针组小鼠大脑神经元每克组织葡萄糖的摄取率均有显著提高(P<0.05)。结论:"通督启神"电针疗法可提高AD模型小鼠的空间学习记忆能力与大脑神经元葡萄糖代谢水平,且疗效优于胆碱酯酶抑制剂多奈哌齐。
Objective: To observe the effects of electro-acupuncture therapy of 'Tongdu Qishen' on spatial learning and memory ability and glucose metabolic level of neurons in the brain of mice with Alzheimer's disease(AD), and evaluate the curative effect of electro-acupuncture therapy of 'Tongdu Qishen' on AD. Methods: Thirty SAMP8 mice were randomly divided into AD group, positive control group and electro-acupuncture therapy group. The other 10 SAMR1 mice served as control group. Mice in electro-acupuncture therapy group received electro-acupuncture stimulation on Baihni (DU20) and Yintang (EX-HN3) for 20min per day with the frequency of electro-acupuncture was 2Hz, and acupuncture at Shuigou (DU26) before electro-acupuncture therapy. Mice in positive group received gavage administration with Donepezil 0.65μg · g^-1. d^-1. Mice in control group and AD group received restriction. After treatment for 15 days, behavioral changes of mice were observed by using Morris water maze, and glucose metabolic level of mice brain was tested by using positron emission tomography scans (PET scans). Results: Compared with AD group, the spatial learning and memory ability and glucose uptake rate of neurons in the brains of mice in electro-acupuncture group were improved significantly (P〈0.05). Compared with positive control group, glucose uptake rate of neurons in the brains of mice in electro-acupuncture group were improved significantly (P〈0.05). Conclusion: Electro-acupuncture therapy of 'Tongdu Qishen' could improve the spatial learning and memory ability and glucose metabolism of brain of mice with AD, and the curative effect is superior to Donepezil.
