摘要
目的:研究芎麻汤药效部位对硝酸甘油所致偏头痛大鼠行为学症状、血浆及脑组织中三叉神经颈髓复合体(TCC)上5-羟色胺(5-HT)的影响,初步探讨其药效部位的作用机制。方法:32只雄性SD大鼠随机分为空白组(A组),模型组(B组),芎麻汤药效部位组(C组),琥珀酸舒马普坦阳性对照组(D组),每组8只;采用皮下注射硝酸甘油(10mg/kg)复制偏头痛模型,观察大鼠挠头、爬笼、咬尾、往返运动,取颈外静脉血采用ELISA检测血浆中5-HT含量,心脏灌注后取大鼠TCC采用免疫组化检测5-HT的阳性表达。结果:芎麻汤药效部位能有效减轻偏头痛大鼠行为学症状,有效降低偏头痛大鼠血浆中5-HT含量,有效降低偏头痛大鼠TCC内5-HT免疫阳性染色光密度值,与模型组比较,差异具有统计学意义(P<0.05,P<0.01)。结论:芎麻汤药效部位能干预硝酸甘油诱导的偏头痛,其机制可能与调节体内5-HT水平有关。
Objective: To study the effects of active extracts of Xiongma Decoction on behavioral symptoms and levels of 5-HT in plasma and trigeminocervical complex (TCC) on animal model of migraine with Nitroglycerin. Methods: Thrity-two SD rats were randomly divided into 4 groups: blank group (group A), model group (group B), Xiongma Decoction group (group C), and sumatriptan group (group D). The migraine model was established by giving hypodermic injection of 10mg/kg Nitroglycerin, the scratching, climbing, biting, back and forth movement were observed, the content of 5-HT on plasma and the expression of 5-HT on TCC were detected by ELISA and stry method. Results: Compared with the model group, Xiongma Decoction could effectively reduce the behavioural symptoms of migraine rat, the content of 5-HT on plasma and positive immunostaining IOD of 5-HT on TCC in Xiongma Decoction group were decreased significantly (P〈0.05, P〈0.01). Conculusion: Xiongma Decoction is effective on the Migraine rat induced by nitroglycerin, the mechanism may be related to adjusting the level of 5-HT.
